Characterization of the stemness potency of mammospheres isolated from the breast cancer cell lines

Abstract: Stemness phenotype mammospheres established from cell lines and tissues taken from autopsy can be used to test and to identify the most sensitive drugs for chemotherapy. Therefore, the aim of the present study was isolation and characterization of cancer stem cells derived from MCF7, MDA-MB231, and SKBR3 breast cancer cell lines to demonstrate the stemness phenotypes of mammospheres generated for further their applications in therapeutic approaches. In this study, two luminal subtypes of cell lines, MCF7 and S… Show more

“…To mimic tumor microenvironments and to enrich stem cell-like cells, a robust in vitro three-dimensional colorectal cancer spheroidal culture was developed and described in this work. In line with other works [49,50], the spheroidal culture showed passage-dependent phenotypic and molecular changes and displayed multiple Predicted complementary binding site of circRNA (red) against the putative target miRNA (green).…”

“…There are two types (ductal and lobular) and multiple subtypes [luminal A and B, basal-like triple negative breast cancer (TNBC), HER2+, and claudin-low] of BC that are classified by molecular and histological phenotypes. Each type and subtype of BC are categorized to non-invasive and invasive (2,(4)(5)(6)(7). A population of cancer cells in BC tissues makes the treatment of BC obscure.…”

“…Hence, our subsequent screening compared the effect of each drug on mammosphere formation, a functional estimation of the number of CSCs, 15,52 versus its effect on the viability of adherent cultures, which are enriched in nonstem cells. 53,54 Pranlukast decreased the number of mammospheres with limited cytotoxicity to adherent cultures, supporting that it selectively targets the CSC pool. Pranlukast binding mode to CD49f was further characterized by MD simulations.…”
