Chemical Methods for the Simultaneous Quantitation of Metabolites and Proteins from Single Cells

Abstract: We describe chemical approaches for integrated metabolic and proteomic assays from single cells. Quantitative assays for intracellular metabolites, including glucose uptake and 3 other species, are designed as surface-competitive binding assays with fluorescence readouts. This enables integration into a micro-array format with functional protein immunoassays, all of which are incorporated into the microchambers of a single-cell barcode chip (SCBC). Using the SCBC, we interrogate the response of human-derived g… Show more

“…The fabrication of the SCBC devices and the protocol of the single cell proteomic assays were extensively discussed in our previous publications (13,14). Briefly, the DNA microarrays at each microchamber were converted to antibody microarrays by flowing the DNA-antibody conjugate cocktail solution immediately prior to use.…”

“…The challenge to do so is because of the targets’ chemical diversity, each requiring different preparation and detection protocols. Xue et al 258 developed a “single cell barcode chip” (SCBS) for quantitative, single cell protein and metabolite measurement. The SCBS chip houses 310, 1.5 nL chambers, which are used for loading and lysing a single cell via a valve to an adjoining lysis buffer compartment.…”

“…Additional reports in which integrated measurements of genes and transcripts 23 , limited numbers of proteins, transcripts 24, 25 and genes 26 , and panels of proteins and metabolites, 27 from single cells have also appeared. Microfluidic methods permit molecular analysis to be correlated with measurements of specific cellular functions (such as motility), or allow the analysis of defined, small populations of cells (i.e.…”
