ChemInform Abstract: First Total Synthesis of Taxol. Part 2. Completion of the C and D Rings.

Abstract: First Total Synthesis of Taxol. Part 2. Completion of the C and D Rings.-The total synthesis of taxol starting with the intermediate (I) ( preparation described in the preceding paper) requires completion of the C ring, introduction of the oxetane ring D and oxidation at C(9) followed by adjustment of the C(9) and C(10) regio-and stereochemistry. The overall yield of taxol (II) from the diol (I) of the preceding paper is ca. 4-5%. -(HOLTON, R. A.; KIM, H.-B.; SOMOZA, C.; LIANG, F.; BIEDIGER, R. J.; BOATMAN, P.… Show more

“…The first successful synthesis originates from Robert Holton. [313,314] Almost simultaneously, Kyriacos Costa ("K. C. ") Nicolaou [315,316] published a total synthesis, and a little later Samuel Danishefsky as well. [317,318] Additional total syntheses were published by Wender, Kuwajima and Mukaiyama.…”

“…To date, seven methods was used in description the total synthesis of taxol [94][95][96][97][98][99][100][101][102][103]. Nicolaou and his co-workers were reported total synthesis of taxol and in 1994 [104][105][106][107].…”
