Chiral Pharmaceuticals

Abstract: The implication of chirality in pharmaceuticals is briefly reviewed, presenting the basic concepts and the chiral recognition phenomenon in biological systems. The importance to recognize enantiomers of chiral pharmaceuticals, both in pharmacology and environmental fields, is emphasized and many examples were selected to demonstrate the significance of chirality in both areas. Methodologies to obtain single enantiomers and to control the enantiomeric fraction associated with enantioseparation of chiral pharmac… Show more

“…Despite the fact that natural compounds possess pharmacological applications, their structures are limited to their production, and sometimes, comprise high levels of complexity, making them difficult to extract and purify, and even harder to synthesize. SAR studies are meant to determine the important moieties of natural compounds in order to improve their pharmacological/biological properties with smaller and simple molecules [88,89,90].…”

“…The chiral analysis by HPLC is based on a formation of transient diastereomeric complexes between the amino acids present in the hydrolysates and the chiral stationary phase (CSP) employed, being the less stable complex the first to elute [ 32 ]. There are several types of CSPs, such as polysaccharide-based, Pirkle-type, protein-based, macrocyclic antibiotic-based, crown ether-based, ligand exchange type, among others [ 33 , 34 , 35 ]; however, the last three types are the most used for the separation of primary amine-containing compounds and amino acids [ 36 , 37 ].…”

“…In chiral drugs, knowledge about the stereochemistry is crucial and accompanies several steps of drug discovery and development. Thus, it can influence absorption, distribution, metabolism and excretion, as well as the molecular mechanism of action, through the phenomenon of chiral molecular recognition [2,4].…”
