Chitosan/tripolyphosphate nanoparticle as elastase inhibitory peptide carrier: characterization and its in vitro release study

Abstract: This study was aimed at the preparation and evaluation of walnut meal derived elastase inhibitory peptide (FFVPF) loaded in chitosan-tripolyphosphate (CS-TPP) nanoparticles (NPs). It was shown that the maximum encapsulation e ciency of FFVPF could reach 94.58 ± 0.23%. TEM microphotographs, polydispersity index and zeta-sizer reports indicated that FFVPF-loaded CS-TPP NPs were in nanometric range and were spherical, discrete and uniform in size with PDI less than 0.3. FTIR analysis indicated that the peptides i… Show more

“…The FTIR spectra of the TPP-Chitosan and TPP-Chitosan-ZnO NPs are shown in Figure . A typical band at 3362 cm –1 can be attributed to the −OH stretching vibration, which overlaps the stretching band of −NH, making it a broad band, as previously observed. , The absorption bands centered around 1980, 1640, 1550, and 1375 cm –1 and present for both TPP-Chitosan and TPP-Chitosan-ZnO could correspond to symmetric and asymmetric CO stretching, respectively. , Besides, the peak at about 1050 cm –1 indicates that the phosphate moiety of TPP is connected to the amine group of chitosan based on electrostatic interaction, which agrees with a previous report . Following incorporation of ZnO NPs into TPP-Chitosan NPs, the peak at 1050 cm –1 was slightly shifted, suggesting a likely interaction of dissolved Zn ions with a −OH group in the TPP-Chitosan.…”

Furthermore, two new absorption peaks at around 465 and 450 cm −1 were noticed, which correspond to the stretching mode of ZnO and align with observations from previous studies.…”
