Chlorophyll Determination in Intact Tissues Using N,N-Dimethylformamide

Abstract: Photosynthetic pigments from etiolated cucumber (Cucumis sativus var. Beit Alpha improved, Hazera Co., Gedera) cotyledons were extracted by direct immersion of the intact cotyledons into the solvent N,N-dimethylformamide (DMF). The solvent is especially efficient when pigment concentration is low; time and tools are saved and the loss of pigment that usually occurs in more complicated extraction procedures is prevented. The specific absorption coefficient of chlorophyll a in DMF was also deternined.Problems in… Show more

“…Chlorophyll a (Chl a), chlorophyll b (Chl b), and total chlorophyll were determined by placing about 15 mg of fresh leaf in 7 ml of N-N dimethylformamide (DMF) for 24 h in the dark at 48C for extraction (Moran & Porath 1980). The absorbance of the extracts was measured spectrophotometrically at 664 and 647 nm and chlorophyll was determined following the equation proposed by Inskeep and Bloom (1985).…”
