Chlorophyll Fluorescence Video Imaging: A Versatile Tool for Identifying Factors Related to Photosynthesis

Abstract: Measurements of chlorophyll fluorescence provide an elegant and non-invasive means of probing the dynamics of photosynthesis. Advances in video imaging of chlorophyll fluorescence have now made it possible to study photosynthesis at all levels from individual cells to entire crop populations. Since the technology delivers quantitative data, is easily scaled up and can be readily combined with other approaches, it has become a powerful phenotyping tool for the identification of factors relevant to photosynthesi… Show more

“…One of the most reliable markers of this process is decreased photosystem II (PS II) activity [98]. Both relative chlorophyll content and PS II efficiency can be easily quantified with pulse amplitude modulation (PAM) fluorometry [99,100]. Thereby, the ratio of minimum (background) and potentially maximum chlorophyll fluorescence (Fv/Fm) is interpreted as the maximum of PS II photochemical activity and might be considered as a reliable marker of PS II photoinhibition and one of the most important indicators of drought stress [101].…”

“…The invention of video imaging systems was an important breakthrough in the field of chlorophyll fluorescence analysis ( Omasa et al, 1987 ; Fenton and Crofts, 1990 ). In fact, this technique not only allowed to spatially examine the heterogeneity within a sample, but it also made possible to evaluate those changes simultaneously on a wide number of samples, making the CFI more popular as diagnostic ( Baker, 2008 ), screening ( Barbagallo et al, 2003 ) and phenotyping tool ( Rühle et al, 2018 ; Pérez-Bueno et al, 2019 ).…”

“…Він характеризує період теплової адаптації листка. При малому значенні періоду адаптації перехід має більшу крутість, а при великих значеннях він більш повільний (Rühle et al, 2018). Параметр F m вказує на найвищий рівень флуоресценції, який реєструється на індукційній кривій у вигляді максимуму.…”
