Cholesterol Esterification Inhibition Suppresses Prostate Cancer Metastasis by Impairing the Wnt/β-catenin Pathway

Abstract: Dysregulation of cholesterol is a common characteristic of human cancers including prostate cancer. This study observed an aberrant accumulation of cholesteryl ester in metastatic lesions using Raman spectroscopic analysis of lipid droplets in human prostate cancer patient tissues. Inhibition of cholesterol esterification in prostate cancer cells significantly suppresses the development and growth of metastatic cancer lesions in both orthotopic and intracardiac injection mouse models. Gene expression profiling… Show more

“…In an imatinib-resistant chronic myelogenous leukaemia cell line, inhibition of CE by the ACAT1 inhibitor avasimibe suppresses tumour growth and subsequently restores imatinib sensitivity by downregulating MAPK signaling 110 . In prostate cancer, avasimibe treatment impairs the Wnt-β-catenin pathway and thus suppresses metastasis 111 . Application of avasimin, a nanomedicine version of avasimibe encapsulated with human serum albumin 112 , specifically induces cancer cell apoptosis and is effective in tumour xenograft models.…”

“…CE inhibition by the ACAT1 inhibitor avasimibe suppresses tumor growth and restores imatinib sensitivity by downregulating MAPK signaling in imatinib-resistant myeloid leukemia [ 92 ]. In prostate cancer, avasimibe inhibits metastasis by blocking the Wnt-β-catenin pathway [ 93 ]. Avasimin (human albumin-capsulated avasimibe) specifically induces tumor apoptosis in a tumor xenograft model [ 94 ].…”

“…The expression was prominent in the tumor masses that developed following the subcutaneous injection of PC3 cells. The intracardiac injection in mice introduced the tumor cells directly into the circulation, thus facilitating the survival and growth of prostate cancer cells in the other organs (31). We used this mouse model to evaluate the effect of Tmem64 in metastatic prostate cancer.…”
