Background: Rapid, reliable Candida species (spp.) identification is crucial because it enables prompt and effective antifungal treatment. Unfortunately this is difficult due to a lack of knowledge, adequate reagents, and equipments, particularly in countries with limited resources. Objectives: The aim of this study was to assess the effectiveness of rapid commercial methods in identifying Candida spp. isolated from critically ill patients. Methodology: Various clinical specimens were collected from critically ill patients admitted to the Intensive Care Units of Mansoura University Hospitals, and processed by numerous mycological techniques. Gram stained, and lactophenol cotton blue stained films, as well, germ tube test, and corn meal agar with 1% Tween 80 were used to identify the isolated Candida spp. Three commercial methods were used for spp. identification; CHROMagar TM Candida medium, API 20 C AUX and Vitek-2 Compact automated system. In addition, all Candida isolates were subjected to species-specific multiplex PCR as the reference method. Results: A total of 52 Candida strains were identified where C. albicans was the predominant spp. (53.8%) followed by C. parapsilolsis (17.3%), C. glabrata (13.5%), C. dubliniensis (5.8%), C. tropicalis (5.8%), C. auris (1.9%) and C. krusei (1.9%). Both CHROMagar TM Candida medium and Vitek-2 Compact automated system exhibited high sensitivity and specificity rates in identification of various Candida spp. as compared to multiplex PCR. Conclusion: This study demonstrated a good performance of both CHROMagar TM Candida medium and Vitek-2 Compact automated system; however, CHROMagar TM Candida medium may be an easier and more cost-effective choice to identify Candida spp., mainly in areas with restricted resources.

Candida species. is a member of the mucous membrane of the normal flora, gastrointestinal tract and skin. They are endogenous opportunistic pathogens that cause secondary infections with underlying immunocompromised condition. Candidiasis is a common fungal infection in human. This study was aimed to isolation and identification of Candida using germ tube, chlamydospore formation, and chromogenic agar testes and to asses antifungal susceptibility to some isolates. A total of 180 samples of skin swabs and skin particles, nail and hair were collected from patients in Duhok Province Hospitals, outpatient and a number of private clinic dermatologists in Duhok city. A Total of 63 Candida isolates was detected in this study. These isolates were subjected to germ tube test, chlamydospores formation and inoculation on commercially available CHROMagar. Candida albicans was the main yeast species isolated followed by C. krusei, C. tropicalis and C. glabrata CHROMO agar is a convenient and very fast method of identifications of Candida species even in resources poor settings. The result of antifungal sensitivity test revealed that itraconazole is the main active antifungal against all Candida species with the minimum inhibition concentration (MIC) of 25 μg /ml. followed by terbinafine, fluconazole and griseofulvin.

Mayalar, gıdaların kalite ve güvenliğini etkileyen gıda endüstrisi için oldukça önemli mikroorganizmalar olup gıdalarda bozulmalara yol açabilirler. Candida türleri hastane enfeksiyonlarında önemli bir yere sahip iken son yıllarda gıdalar vasıtasıyla da insanlara bulaşabilmektedir. Bu çalışma ile halk pazarlarından toplanan peynirlerde patojen Candida türlerinin varlığının araştırılması amaçlandı. Analiz edilen 100 adet peynir örneğinin 84 (%84)'ünde patojen Candida spp. pozitif olarak bulundu ancak hiçbirinde C. albicans saptanamadı. Bu çalışmada API 20 C AUX Test Kit'i ile 103 adet izolatın tür ayrımı yapıldı. Elde edilen test kit sonuçlara göre izolatların %14'ü C. krusei, %5'i C. tropicalis olarak identifiye edildi. Gıda örneklerinin mikrobiyolojik incelemesinde toplam maya ve küf sayısı ile birlikte patojen mayaların varlığına yönelik analizlerin de yapılması toplum sağlığı açısından yararlı olacağı sonucuna varıldı.
