Chromatographic Method for Determination of the Amino Acid Content in Dioscorea bulbifera L. Tubers by RP-HPLC

Abstract: Background: The present study was carried out for determination of amino acid content in tubers of Dioscorea bulbifera using reverse-phase high-performance liquid chromatography. Methods: The method involved the vapor phase hydrolysis of the sample, automated derivatisation of the amino acids with the aid of AccQ-Fluor reagent kit, separated on a high performance liquid chromatography equipped with photo diode array (HPLC-PDA) at 254 nm having column temperature of 37 ºC. Results: The proportional molar concen… Show more

“…Its simplicity, ecofriendly and cost-effectiveness are the advantages of using GC instead of other techniques for the quantification of pesticides from natural sources. In continuation of our previous study (Bhat et al, 2019;Dangroo et al, 2014;Dangroo, Singh, et al, 2016;Dar et al, 2014Dar et al, , 2015Dar et al, , 2016Fayaz et al, 2018;Khan et al, 2013;Singh et al, 2018) on the application of chromatography for fingerprinting and chromatographic method development to assure the quality of vegetables/medicinal plants, a validated analytical method has been developed for the quantification of cypermethrin. Besides the development of a validated method, risk assessment and determination of the dissipation behaviour, persistence and half-life period of cypermethrin on green pea were carried out.…”

“…27,[55][56][57] The instrumental methods of analysis are based upon the measurement of some physical properties of a substance to determine its chemical composition, quality, and quantification. 25,56,58 Presently, many sophisticated techniques have emerged, among which spectrophotometric and chromatographic techniques are being most widely used for the sample analysis. 48,59,60 The main objective of these methods is the analysis of the drug/marker which is of interest alone in the matrix of the excipients, additive, degradation products, impurities and so forth, and also other drugs in case of the combination products.…”
