Chromosomal damage in peripheral blood lymphocytes of newly diagnosed cancer patients and healthy controls

Abstract: The frequencies of aberrant cells (ACs) and CAs were significantly higher in patients (2.38 +/- 1.56 and 2.53 +/- 1.69, respectively) as compared with controls (1.81 +/- 1.31 and 1.94 +/- 1.47, respectively, P < 0.01). The percentual difference in chromatid-type aberrations (CTAs) between patients and controls was moderately significant (1.37 +/- 1.20 and 1.11 +/- 0.99, respectively, P Show more

“…This has been shown for diseases like Irritable Bowel Syndrome and diabetes (Collins et al, 1998b;Najafzadeh et al, 2009;Wyatt, 2006). It also confirms findings of Vodicka et al who observed increased chromosomal damage in lymphocytes of newly diagnosed cancer patients compared with healthy controls (Vodicka et al, 2010). Similar observations of a higher baseline DNA damage were made for head and neck squamous cell carcinoma patients (Palyvoda et al, 2003) and breast cancer patients (Rajeswari et al, 2000;Smith et al, 2003) in addition to higher levels of cytogenetic damage (Palyvoda et al, 2003).…”

“…Healthy individuals carrying high EPHX1 activity genotypes were recently found to have a decreased frequency of nonspecific chromosomal aberrations (Hemminki et al 2015). As an increased frequency of chromosomal aberrations predicts cancer risk (Vodicka et al 2010) this study provides a biological link between genetic susceptibility and genotoxicity with potential implication for carcinogenesis.…”

“…This is an important issue, since the abovementioned compounds are classified as proven (formaldehyde and cyclophosphamide, for instance) or potential human carcinogens (13,29,30) and the level of CA is considered to be predictive of cancer risk (31)(32)(33).…”

“…The methodology for cytogenetic analysis has been described earlier (33,34) and was in accordance with existing guidelines (35). Briefly, peripheral blood lymphocytes stimulated to grow by phytohemagglutinin were cultured for 48 hours.…”
