BackgroundHairy beggar-ticks (Bidens pilosa L.) is a common invasive plant in tropical and subtropical regions. The Flora of Taiwan listed three varieties of B. pilosa in Taiwan, var. minor, var. pilosa and var. radiata. Among the three varieties, var. radiata was the most recently, in 1970s, introduced into Taiwan. However, after its introduction into Taiwan, var. radiata has become dominant over the other two varieties and is considered a serious invasive plant in lowland of Taiwan. Our previous study showed that var. radiata is self-incompatible and the other two varieties are self-fertile. Could it be possible that different chromosome numbers contribute to the different breeding systems of these three varieties? In addition, the heterogeneities of traits of var. radiata were found higher than those of var. minor and var. pilosa. Is the phenomenon resulting from the hybridization between var. radiata with other varieties? We counted chromosome numbers of populations of these three varieties distributed in Taiwan and conducted hand pollination treatment between var. radiata (as pollen receiver) and var. minor or var. pilosa (as pollen donor) to provide answer for the aforementioned questions.ResultsNo difference was found in chromosome numbers among populations of the same variety. Forty-eight chromosomes (2n = 48) were counted for var. radiata while 72 (2n = 72) chromosomes for var. minor and var. pilosa. Therefore, var. radiata is tetraploid and var. minor and var. pilosa are hexaploid. No successful hybridization was found between var. radiata and var. minor or between var. radiata and var. pilosa.ConclusionsThis study provided the evidence that the invasive plant (B. pilosa var. radiata) has different chromosome numbers from the other two varieties and is unlikely to hybridize with the other two varieties.

BackgroundPressurized hot water extraction (PHWE) technique has recently gain much attention for the extraction of biologically active compounds from plant tissues for analytical purposes, due to the limited use of organic solvents, its cost-effectiveness, ease-of-use and efficiency. An increase in temperature results in higher yields, however, issues with degradation of some metabolites (e.g. tartrate esters) when PHWE is conditioned at elevated temperatures has greatly limited its use. In this study, we considered possibilities of optimizing PHWE of some specific functional metabolites from Bidens pilosa using solvent compositions of 0, 20, 40 and 60 % methanol and a temperature profile of 50, 100 and 150 °C.ResultsThe extracts obtained were analyzed using UPLC-qTOF-MS/MS and the results showed that both temperature and solvent composition were critical for efficient recovery of target metabolites, i.e., dicaffeoylquinic acid (diCQA) and chicoric acid (CA), which are known to possess anti-HIV properties. It was also possible to extract different isomers (possibly cis-geometrical isomers) of these molecules. Significantly differential (p ≤ 0.05) recovery patterns corresponding to the extraction conditions were observed as recovery increased with increase in methanol composition as well as temperature. The major compounds recovered in descending order were 3,5-diCQA with relative peak intensity of 204.23 ± 3.16 extracted at 50 °C and 60 % methanol; chicoric acid (141.00 ± 3.55) at 50 °C and 60 % methanol; 4,5-diCQA (108.05 ± 4.76) at 150 °C and 0 % methanol; 3,4-diCQA (53.04 ± 13.49) at 150 °C and 0 % methanol; chicoric acid isomer (40.01 ± 1.14) at 150 °C and 20 % methanol; and cis-3,5-diCQA (12.07 ± 5.54) at 100 °C and 60 % methanol. Fitting the central composite design response surface model to our data generated models that fit the data well with R2 values ranging from 0.57 to 0.87. Accordingly, it was possible to observe on the response surface plots the effects of temperature and solvent composition on the recovery patterns of these metabolites as well as to establish the optimum extraction conditions. Furthermore, the pareto charts revealed that methanol composition had a stronger effect on extraction yield than temperature.ConclusionUsing methanol as a co-solvent resulted in significantly higher (p ≤ 0.05) even at temperatures as low as 50 °C, thus undermining the limitation of thermal degradation at higher temperatures during PHWE.

Background The Coreopsideae tribe, a subset of the Asteraceae family, encompasses economically vital genera like Dahlia, Cosmos, and Bidens, which are widely employed in medicine, horticulture, ecology, and food applications. Nevertheless, the lack of reference genomes hinders evolutionary and biological investigations in this tribe. Results Here, we present 3 haplotype-resolved chromosome-level reference genomes of the tribe Coreopsideae, including 2 popular flowering plants (Dahlia pinnata and Cosmos bipinnatus) and 1 invasive weed plant (Bidens alba), with assembled genome sizes 3.93 G, 1.02 G, and 1.87 G, respectively. We found that Gypsy transposable elements contribute mostly to the larger genome size of D. pinnata, and multiple chromosome rearrangements have occurred in tribe Coreopsideae. Besides the shared whole-genome duplication (WGD-2) in the Heliantheae alliance, our analyses showed that D. pinnata and B. alba each underwent an independent recent WGD-3 event: in D. pinnata, it is more likely to be a self-WGD, while in B. alba, it is from the hybridization of 2 ancestor species. Further, we identified key genes in the inulin metabolic pathway and found that the pseudogenization of 1-FEH1 and 1-FEH2 genes in D. pinnata and the deletion of 3 key residues of 1-FFT proteins in C. bipinnatus and B. alba may probably explain why D. pinnata produces much more inulin than the other 2 plants. Conclusions Collectively, the genomic resources for the Coreopsideae tribe will promote phylogenomics in Asteraceae plants, facilitate ornamental molecular breeding improvements and inulin production, and help prevent invasive weeds.
