Chronic atrial fibrillation causes left ventricular dysfunction in dogs but not goats: experience with dogs, goats, and pigs

Abstract: Structural remodeling in chronic atrial fibrillation (AF) occurs over weeks to months. To study the electrophysiological, structural, and functional changes that occur in chronic AF, the selection of the best animal model is critical. AF was induced by rapid atrial pacing (50-Hz stimulation every other second) in pigs (n ϭ 4), dogs (n ϭ 8), and goats (n ϭ 9). Animals underwent MRIs at baseline and 6 mo to evaluate left ventricular (LV) ejection fraction (EF). Dogs were given metoprolol (50 -100 mg po bid) and … Show more

“…We analysed a simpler condition of increased tissue anisotropy, which can be linked to diffuse interstitial fibrosis. This is representative of both dog models of AF and of 70% of AF patients who do not develop severe patchy fibrosis [45,46]. This modelling approach also enables a separation of purely electrophysiological effects from those related to the presence of patchy fibrosis.…”

“…However, the mechanism involved in the development of nonischemic LV-LGE patterns in AF patients is still unclear. This could result from LV remodeling associated with chronic AF (24) or aging (25), or from another multifactorial, genetic process.…”

“…Procedures for the pacemaker implantation and programming have been described in detail previously . In brief, mixed breed goats were fasted 12–24 hours prior to surgery, anesthetized with propofol (5–8 mg/kg iv), and maintained with inhaled isoflurane dosed to effect (1.5–4%) in inspired oxygen.…”
