1 These authors contributed equally to this workMaturing of a messenger RNA (mRNA) is a multi-way process producing mRNA variants through diverse splicing events, alternative polyadenylation, RNA editing, etc. Studying posttranscriptional processing of human cold-inducible RNA binding protein (CIRBP), we discovered yet another mechanism that could be added to this list. We named it excising, and it consists of low-accuracy post-splicing deletion of sequence regions of variable length. The main features of the excising process and putative members of corresponding multiprotein machinery were described with a series of cloning vectors and RNApulldown assay. Our results highlight a possible role of U-rich stretches and the proteins targeting such motifs in the discovered process. The discovered mechanism suggests the potential translation of 3'-untranslated regions, which may be an adjuvant way of CIRBP activity inhibition or generation of structural and functional diversity.Alternative splicing | RNA editing | RNA modification | RNA metabolism Correspondence: reshetnikov@fbb.msu.ru

The precision evaluation of prognosis is crucial for clinical treatment decision of bladder cancer (BCa). Therefore, establishing an effective prognostic model for BCa has significant clinical implications. We performed WGCNA and DEG screening to initially identify the candidate genes. The candidate genes were applied to construct a LASSO Cox regression analysis model. The effectiveness and accuracy of the prognostic model were tested by internal/external validation and pan-cancer validation and time-dependent ROC. Additionally, a nomogram based on the parameter selected from univariate and multivariate cox regression analysis was constructed.Eight genes were eventually screened out as progression-related differentially expressed candidates in BCa. LASSO Cox regression analysis identified 3 genes to build up the outcome model in E-MTAB-4321 and the outcome model had good performance in predicting patient progress free survival of BCa patients in discovery and test set. Subsequently, another three datasets also have a good predictive value for BCa patients' OS and DFS. Time-dependent ROC indicated an ideal predictive accuracy of the outcome model. Meanwhile, the nomogram showed a good performance and clinical utility. In addition, the prognostic model also exhibits good performance in pan-cancer patients. Our outcome model was the first prognosis model for human bladder cancer progression prediction via integrative bioinformatics analysis, which may aid in clinical decision-making. Background: Dysregulation of RNA-binding proteins (RBPs) playsan important role in controlling processes in cancer development.However, the function of RBPs has not been thoroughly and systematically documented in head and neck cancer.We aim to explore the role of RPB in the pathogenesis of HNSC.Methods: We obtained HNSC gene expression data and corresponding clinical information from The Cancer Genome Atlas (TCGA) and the GEO databases, andidentified aberrantly expressed RBPs between tumors and normal tissues.Meanwhile, we performed a series of bioinformatics to explore the function and prognostic value of these RBPs.Results: A total of 249 abnormally expressed RBPs were identified, including 101 down-regulated RBPs and 148 up-regulated RBPs.Using least absolute shrinkage and selection operator (LASSO) and univariate Cox regression analysis, the fifteen RPBs were identified as hub genes. With the fifteen RPBS, the prognostic prediction model was constructed.Further analysis showed that the high-risk score of the patients expressed a better survival outcome. The prediction model was validated in another HNSC dataset, and similar findings were observed. Conclusions: Our results provide novel insights into the pathogenesis of HNSC. The fifteen RBP gene signature exhibited the predictive value of moderate HNSC prognosis, and have potential application value in clinical decision-making and individualized treatment.
