Circular RNA and Its Roles in the Occurrence, Development, Diagnosis of Cancer

Abstract: Circular RNAs (circRNAs) are non-coding single-stranded covalently closed circular RNA, mainly produced by reverse splicing of exons of precursor mRNAs (pre-mRNAs). The characteristics of high abundance, strong specificity, and good stability of circRNAs have been discovered. A large number of studies have reported its various functions and mechanisms in biological events, such as the occurrence and development of cancer. In this review, we focus on the classification, characterization, biogenesis, functions o… Show more

“…Non-coding RNAs accounted for approximately 60% of the transcriptional output of human cells and had been shown to regulate a variety of cellular processes under physiological and pathological conditions ( 13 , 52 ). RNA polymerase II was also required by cancer-related genes in eukaryotes nucleus to produce non-coding RNAs such as circRNAs and this synergistic process was precisely regulated by multiple factors ( 53 ).…”

“…CircRNAs are closed-loop molecules with unique structures that are widely distributed in animal and plant cells and regulate gene expression. They exist independently of proteins without being affected by exonucleases ( Yue Zhang et al, 2022 ). Because of their low abundance and the limitation of detection technology, circRNAs were initially ignored as abnormal products of RNA splicing ( Wen et al, 2022 ).…”

“…Circ-LARP1B/miR-515-5p/TPX2 microtubule nucleation factor regulatory axis is involved in the malignant progression of CSCC by promoting cell viability, colony formation, migration, invasion, cell cycle progression, and glycolysis and inhibiting apoptosis ( Yan Wang et al, 2022 ). Circ_0001821 plays a pro-oncogenic role in CSCC development by regulating the miR-148a-3p/EGFR signaling axis and PI3K/Akt pathway ( Yue Zhang et al, 2022 ).…”

“…Circular RNAs (circRNAs) are mainly produced by the reverse splicing of exons of precursor mRNAs. The functions of circRNAs are mainly ceRNA or miRNA sponging, binding with proteins, regulation of precursor mRNAs (pre-mRNA) splicing, regulation of parental gene expression, and potential translation templates for proteins or peptides ( Yan and Bu, 2021 ; Zhang Y. et al, 2022 ). MiRNAs, lncRNAs, and circRNAs may have other ways to regulate the expression of mRNAs or proteins jointly or competitively.…”
