Circular RNA HIPK3 induces cell proliferation and inhibits apoptosis in non-small cell lung cancer through sponging miR-149

Abstract: Circular RNAs (circRNAs) are a class of endogenous noncoding RNAs that are demonstrated to be potent regulators in the development of various types of human cancers, including non-small cell lung cancer (NSCLC). In the present study, the level of circRNA-HIPK3 were measured by Taq-man based quantitative real-time PCR (qRT-PCR) analysis in both NSCLC patient specimens and cells, which showed that circRNA-HIPK3 was upregulated in both NSCLC tissues and cell lines. Cell counting kit-8 (CCK-8), migration and flow-… Show more

“…Non-invasive prognostic biomarkers are needed for the diagnosis of the advanced NSCLC patients with different histological types [18]. With the advance in the study of the molecular basis about lung cancer progression, targeted therapies have been used for NSCLC treatment [19]. CircRNAs are a group of non-coding RNAs with a stable closed-loop structure which prevents them from decomposing by the enzymes [20].…”

“…circHIPK3 (hsa_circ 0000284) is a circular RNA derived from the second exon of the HIPK3 gene. circHIPK3 is highly expressed in liver, brain, and lung (13)(14)(15) and has shown an important role in the regulation of cell survival, oxidative damage, and inflammation (16)(17)(18). We note that circHIPK3 is also expressed in pancreatic tissue (19).…”

“…Even though it is not deniable that CK2 plays a central role in cancer diseases it is likely that antiproliferative effects of silmitasertib (1) are also a consequence of inhibiting off-targets [29]. Main off-targets of silmitasertib (1) reported to have antiproliferative effects are for example the cyclin-dependent kinase (CDK1) [43,44], PIM-1 protooncogene, serine/threonine kinase (PIM1) [45], Homeodomain-interacting protein kinase 3 (HIPK3) [46,47], TANK-binding kinase 1 (TBK1) [48] or FMS-like tyrosine kinase 3 (FLT3) [49,50]. More recently some more selective CK2 inhibitors have been reported, including GO289 by Oshima et al [51] or an allosteric inhibitor CAM4066 by Brear et al [18].…”
