“…sulin resistance by interacting with phosphatidylinositol 5-phosphate 4-kinase type-2 beta (PIP4k2β) [9], yet systemic MANF overexpression improved glucose tolerance [10]. In contrast to our findings, Wu et al found a positive correlation between circulating MANF with insulin resistance in patients with prediabetes, whereas circulating MANF was not related to insulin resistance in T2DM patients even they had severer insulin resistance than prediabetic patients [11]. Their populations had obviously glucose dysmetabolism and pancreatic β-cell failure, while our participants were mainly characterized by hyperinsulinemia and hyperandrogenism.…”

“…Previous data suggest an association between MANF and insulin resistance [9][10][11]. However, human data have not been consistent, and they did not use the EHC, the golden standard for evaluating insulin resistance [15,16].…”

“…Although MANF was not completely deleted from all pancreatic b-cells in MIP-1Cre ERT ::Manf fl/fl (TMX) mice, we demonstrate that lack of MANF in the adult b-cells results in reduced expression of b-cell-specific genes and poor b-cell survival and proliferation, leading to hyperglycemia and diabetes. The above results strongly argue against a systemic effect of circulating MANF in rescuing b-cells and clearly demonstrates that local expression of MANF is required to maintain the b-cell phenotype in mice (56,57). Furthermore, MANF expression in pancreatic exocrine acinar cells did not protect MANF-deficient adult b-cells.…”

“…26,27 Studies indicated that activation of the central nerve system GLP-1 receptor improves T2DM via glucose metabolism dependent inhibition of central nerve system AMPK. 28 AMPK activation accelerate fatty acid oxidation and glucose uptake, suppresses triglycerides production in skeletal muscle, which is necessary for the body to maintain the balance of blood glucose. 29 In this study, AMPK and p-AMPK were examined to understand whether Ex-4 improves glucose metabolism by increasing phosphorylation of AMPK.…”
