Circulating MicroRNA Is a Biomarker of Biliary Atresia

Abstract: The lack of reliable noninvasive diagnostic biomarkers of biliary atresia (BA) results in delayed diagnosis and worsened patient outcome. Circulating microRNAs (miRNAs) are a new class of noninvasive biomarkers with encouraging diagnostic utility. In this study we examined the ability of serum miRNAs to distinguish BA from other forms of neonatal hyperbilirubinemia. BA-specific serum miRNAs were identified using a microfluidic array platform and validated in a larger, independent sample set. The miR-200b/429 c… Show more

“…32,122,123 In biliary atresia, circulating levels of the miR-200b/429 cluster are elevated in infants and have promising diagnostic clinical potential. 127 Interestingly, exosomal miRNA expression pattern is unique to liver diseases and varies between grade and stages of liver disease, which emphasizes their potential as diagnostics biomarkers. 128 Moreover, circulating miR-122 could be used as a general marker of liver injury and might have better sensitivity than alanine aminotransferase.…”

“…As in the study conducted in CFLD, serum miRNA profiles have been evaluated using PCR arrays in children with BA versus a cholestatic control group including children diagnosed with non-BA cholestatic diseases such as Alagille syndrome or choledocholithiasis (Table 3) [144]. Eleven miRNAs were found to be differentially expressed between the two groups and nine confirmed by qRT-PCR in independent cohorts.…”

“…In a recent study, serum microRNAs were assessed for their utility as a diagnostic tool to differentiate BA from other forms of neonatal hyperbilirubinemia [71]. A microRNA array was performed on sera from BA patients and age-and sex- matched indeterminate cholestasis controls.…”
