Circulating miR-210 as a diagnostic and prognostic biomarker for colorectal cancer

Abstract: microRNA-210 (miR-210), the master hypoxamir, is overexpressed and generally exhibits oncogenic properties in most human solid tumours, including colorectal cancer (CRC). However, the status of circulating miR-210 in CRC is still unknown. This study aims to assess the clinical significance of circulating miR-210 in CRC. Using (reverse transcription quantitative PCR) RT-qPCR analysis, we compared the expression levels of circulating miR-210 in serum of 268 CRC patients and 102 healthy controls, and found that s… Show more

“…Patients with breast cancer present increased levels of circulating miR-210; thus, circulating miR-210 may be a potential biomarker of tumor presence and therapeutic response in breast cancer (59). Moreover, hypoxia-inducible circulating miR-210 contributes to metastasis in colorectal cancer (CRC) (60), and it has recently been described as a potential non-invasive marker for diagnosis and prognosis of colorectal cancer (CRC) (61). Given the several different roles that miR-210 plays in numerous aspects of cellular function, it is also noteworthy to mention that hypoxia induces K562 and β-thalassemic erythroid progenitor cell differentiation, and this induction is at least in part mediated by miR-210, which, in turn, modulates a linkage between erythropoiesis and hypoxia (62).…”

“…For instance, microRNA 144 could repress insulin signaling during type 2 diabetes mellitus development through suppressing the expression of Insulin Receptor Substrate 1 (IRS1) [26]. Currently, miR-210 has been reported to have significant contributions in various diseases including cardiovascular ischemic diseases [27,28], cancer [29,30], Graves' disease [31], and psoriasis [32]. Moreover, a group of miRNAs including miR-210 were differentially expressed in exosomes from obese adipose tissue macrophages [15].…”

“…Aberrant miRNA expression is associated with the occurrence of cancer in humans, such as colorectal cancer [8] and NSCLC [9]. Recently, miRNA (miR)-210, which is located on chromosome 11p15.5, was found to have multiple roles in various biological processes, such as angiogenesis and regulation of the immune system, by modulating cell proliferation, differentiation, degradation, and necrosis [10], and miR-210 was involved in the myocardial energy metabolism regulating the energy shift in cardiomyocytes during oxidative stress [11]. Deregulation of miR-210 may aberrantly activate oncogenes, deactivate tumor suppressor genes, inhibit DNA repair, and promote angiogenesis, ultimately accelerating the process of carcinogenesis, especially in a hypoxic environment [12].…”
