When breeding crops that utilize nitrogen efficiently, it is important to reveal the regulation of nitrate uptake at the molecular level. We focused on the expression of two nitrate uptake-related genes, NRT2 and NAR2, and nitrate uptake during the nitrate induction. Four rice NRT2s (OsNRT2.1~2.4) and two NAR2s (OsNAR2.1~2.2) were identified in the rice genome database. We analyzed the expression of the genes in the roots and shoots after supply of nitrate with or without ammonium pretreatment by the RT-PCR method. The apparent nitrate uptake was also measured. Differential expression of OsNRT2.1 and OsNRT2.2, which have the same ORF with different 5′-and 3′-UTR, was observed in the seedlings pretreated with ammonium. The apparent nitrate uptake synchronously increased after supply of nitrate with the expression pattern of OsNRT2.1 in roots either with or without ammonium pretreatment. The expression of OsNAR2.1, which is thought to be an activator gene for the function of NRT2, was also coordinated with that of OsNRT2.1. On the other hand, OsNRT2.2 was already expressed after ammonium pretreatment, suggesting that OsNRT2.2 would be transcribed to take up nitrate in flooded soils. The present results indicate that OsNRT2.1, OsNRT2.2 and OsNAR2.1 are candidate genes when breeding for efficient use of nitrate in rice.

In higher plants, two types of nitrate transporters, NRT1 and NRT2, have been identified. In Arabidopsis, there are 53 NRT1 genes and 7 NRT2 genes. NRT2 are high-affinity nitrate transporters, while most members of the NRT1 family are low-affinity nitrate transporters. The exception is CHL1 (AtNRT1.1), which is a dual-affinity nitrate transporter, its mode of action being switched by phosphorylation and dephosphorylation of threonine 101. Two of the NRT1 genes, CHL1 and AtNRT1.2, and two of the NRT2 genes, AtNRT2.1 and AtNRT2.2, are known to be involved in nitrate uptake. In addition, AtNRT1.4 is required for petiole nitrate storage. On the other hand, some members of the NRT1 family are dipeptide transporters, called PTRs, which transport a broad spectrum of di/tripeptides. In barley, HvPTR1, expressed in the plasma membrane of scutellar epithelial cells, is involved in mobilizing peptides, produced by hydrolysis of endosperm storage protein, to the developing embryo. In higher plants, there is another family of peptide transporters, called oligopeptide transporters (OPTs), which transport tetra/pentapeptides. In addition, some OPTs transport GSH, GSSH, GSH conjugates, phytochelatins, and metals.

Families of nitrogen (N) transporters have been identifi ed in the model plantArabidopsis , and this template is useful for other species. For crops, uptake effi ciency is an important component of nitrogen use efficiency (NUE), which has been shown to vary widely between cultivars and species. The root is the most important organ for acquiring soil nitrogen, which is chiefl y available as NO 3 − , NH 4 + or amino acids. Soil nitrogen form and availability to roots is transient, and the concentrations can rapidly change in response to climatic factors. Although root architecture is undoubtedly a key trait for nitrogen acquisition effi ciency (see Chapter 2 ) and the rhizosphere infl uences availability (Chapter 3 ), the activity of root transporters is important for uptake. Roots have transporter systems to acquire the different nitrogen forms from the soil. Ammonium uptake is achieved by ammonium transporters (AMTs) and amino acids by several different families of transporters.
