Cloning and sequence analysis of the genes encoding the dihydrolipoamide acetyltransferase and dihydrolipoamide dehydrogenase components of the pyruvate dehydrogenase multienzyme complex of Bacillus stearothermophilus

Abstract: A 2641-bp EcoRI fragment of DNA that encodes the C-terminal part of the dihydrolipoyl acetyltransferase (E2) component and the dihydrolipoamide dehydrogenase (E3) component of the pyruvate dehydrogenase complex of Bacillus stearothermophilus has been cloned in Escherichia coli. Its nucleotide sequence was determined. A 705-bp truncated open reading frame was located at the 5' end of the insert which, together with the 588-bp truncated open reading frame at the 3' end of another EcoRI fragment of B. stearotherm… Show more

“…3 Additionally, the possibility of a geneencoded E2 with a single lipoyl domain is not new. The E2 subunits for S. cerevisiae (35), Bacillus stearothermophilus (42), and even A. thaliana plastid (37) PDCs all contain one lipoyl domain. In this report, we extend this list to include maize, A. thaliana, and other, if not all, plant mitochondrial E2s.…”

“…Sequences compared are the potato sequence described in this paper and the sequences from 5. stearothermophilus (Borges et al, 1990) the activity of mtPDC, we measured the activity of the enzyme in mitochondria isolated from cotyledons. These results, shown in Figure 5A, demonstrate that there is a transient increase in mtPDC activity.…”
