Cloning, sequence analysis and heterologous expression of the DNA adenine-(N6) methyltransferase from the human pathogen Actinobacillus actinomycetemcomitans

Abstract: We cloned and sequenced the DNA adenine-N 6 methyltransferase gene of the human pathogen Actinobacillus actinomycetemcomitans (M.AacDAM). Restriction digestion shows that the enzyme methylates adenine in the sequence GATC. Expression of the enzyme in a DAM 3 background shows in vivo activity. A PSI-BLAST search revealed that M.AacDAM is most related to M.HindIV, M.EcoDAM, M.StyDAM, and M.SmaII. The ClustalW alignment shows highly conserved regions in the enzyme characteristic for type a MTases. Phylogenetic tr… Show more

“…Dam was required for virulence in Pasteurella multocida, known to cause bovine respiratory disease (14). Recently, the gene encoding Dam was cloned and sequenced from Actinobacillus actinomycetemcomitans, which is implicated in causing human periodontal diseases (21). Therefore, Dam presents the exciting possibility that it may play a role in the virulence of a broad range of pathogens and, thus, further investigation is merited.…”

“…We identified and characterized the A. actinomycetemcomitans DAM (M.AacDAM) of strain Y4. 17 A DAM-deficient mutant of A. actinomycetemcomitans showed increased levels of extracellular leukotoxin and the reduced capacity to invade epithelial cells. 19 On various epithelial surfaces as well as in the oral cavity structured bacterial biofilms can be pathogenic and induce chemical defense mechanisms or inflammatory responses in epithelial cells.…”

“…have been identified in the oral bacteria Porphyromonas gingivalis and A. actinomycetemcomitans 17,18 and both microorganisms are associated with periodontal diseases. The pattern of DNA methylation is crucial to the regulation of cellular processes, including the expression of various virulence genes.…”

“…DAM (DNA adenine methyltransferase) is one such orphan enzyme, and is prevalent among enteric bacteria, such as E. coli , Salmonella species, Yersinia species and Vibrio cholerae (2). DAM enzymes have also been identified in the oral bacteria Porphyromonas gingivalis (2) and A. actinomycetemcomitans (8). Patterns of DNA methylation are crucial to the regulation of important cellular processes (5, 20, 22, 28), including the expression of various virulence genes.…”
