Cobalt has Enhancing Effect on Extracellular Lipases Isolated from Pseudomonas aeroginosa JCM5962(T)

Abstract: : Present study demonstrates effect of metal ions and inhibitors on the activity of lipase, isolated from P.aeruginosa. Seven different concentrations (0.1mM -20mM) of twelve metal salts and two inhibitors, EDTA and SDS were studied for their effect on lipase. The results demonstrated that ten metal ions, namelyFe +2 andK + were not found to be significantly affecting the enzyme. The enzyme was also tested for two inhibitors, EDTA and SDS; both reduced the enzyme action and SDS showed complete inhibition at hi… Show more

“…As a result, under such conditions the bacterial cell membrane easily transports these ions into the cell and changes its osmotic pressure, which can affect the viability of the bacteria. Researchers have noticed that the activity of P. aeruginosa JCM5962 (T) enzymes and other species of Pseudomonas decreases when the medium has a high Na + ion concentration (Gautam and Azmi 2017;Sachan et al 2017). strategy of adding inoculum progressively was effective in improving the fermentation process.…”

“…The internal osmotic pressure must be higher than in the external environment in order to maintain bacterial growth and viability. If osmotic pressure is too low/high, it could also affect enzyme activity [25,26]. To maintain an acid whey substrates pH, a significant amount of 1 M NaOH is used in samples with a higher proportion of acid whey (Figure 4), influencing lactonase activity and the lactose conversation rate into LBA.…”

“…It was not suprising since most metal ions and detergents are known to negatively impact on activity and stability of lipases [21]. Detergents and metal ions are known to have an influence on the biological function of enzymes, by enhancing or inhibiting their activity via various mechanisms, such as acting as an electron donor or acceptor and forming complexes with limited reactivity [22]. Like any other enzyme, inhibition of lipase activity by these metal ions is caused by inhibition of the catalytic site [19].…”
