Coculture techniques for modeling retinal development and disease, and enabling regenerative medicine

Abstract: Stem cell‐derived retinal organoids offer the opportunity to cure retinal degeneration of wide‐ranging etiology either through the study of in vitro models or the generation of tissue for transplantation. However, despite much work in animals and several human pilot studies, satisfactory therapies have not been developed. Two major challenges for retinal regenerative medicine are (a) physical cell‐cell interactions, which are critical to graft function, are not formed and (b) the host environment does not prov… Show more

“…In the human retina, the RPE cells nourish the neural retina. The microcapillaries between RPE and the neural retina supply oxygen and metabolites 34 . The co-culture technique can enable optimization of regenerative cell therapy for degenerative retinal diseases in vitro 34 .…”

“…The physiological functions of RPE cells included growth factors secretion, the blood-retinal barrier (BRB) maintenance and outer disc membrane phagocytosis [28,29]. Pathological stress, such as aging, would lead to the loss of physiological functions and thus cause advanced pathological changes.…”

“…For this reason, to better reproduce physiological conditions and investigate the role of the interaction between cells, more complex in vitro structures were developed. Thus, RPE cultures can be combined with different retinal cells or an in vitro representation of the choroid capillary bed [ 125 ].…”
