Collagen-Derived Di-Peptide, Prolylhydroxyproline (Pro-Hyp): A New Low Molecular Weight Growth-Initiating Factor for Specific Fibroblasts Associated With Wound Healing

Abstract: Many cells and soluble factors are involved in the wound healing process, which can be divided into inflammatory, proliferative, and remodeling phases. Fibroblasts play a crucial role in wound healing, especially during the proliferative phase, and show heterogeneity depending on lineage, tissue distribution, and extent of differentiation. Fibroblasts from tissue stem cells rather than from healthy tissues infiltrate wounds and proliferate. Some fibroblasts in the wound healing site express the mesenchymal ste… Show more

“…Pro-hydroxyproline was also significantly increased in banded hearts ( Figure 2B ). Given that several of these metabolites are in ECM remodeling, it is likely they contribute to initiation of the fibrotic response and resultant tissue fibrosis ( 34 , 35 , 36 , 37 , 38 , 39 ). The high correlations between the abundance of various metabolites and phenotypic indicators of cardiac remodeling indicate changes in metabolism are not merely consequential, but likely effectors and drivers of disease.…”

“…Generation of tri- and dipeptides was observed in the ear with dermatitis as a result of extensive degradation of ECM, indicating that collagen-derived tri- and dipeptides act as an ECM-associated signal to trigger fibroblast growth for tissue reconstruction [ 56 ]. In fact, Pro-Hyp was incorporated into fibroblasts with mesenchymal stem cell marker, p75NTR, indicating its role in wound healing [ 57 ]. Moreover, tri- and dipeptides containing Hyp induce chemotaxis of fibroblasts by acting on the membrane receptor [ 58 , 59 ] and, especially, Pro-Hyp stimulates cell proliferation and hyaluronic acid synthesis [ 60 ].…”
