Collection, Storage and Protein Extraction Method of Gingival Crevicular Fluid for Proteomic Analysis

Harraa S. Mohammed-Salih,
Hayder F. Saloom

Abstract: Gingival crevicular fluid (GCF) may reflect the events associated with orthodontic tooth movement. Attempts have been conducted to identify biomarkers reflecting optimum orthodontic force, unwanted sequallea (i.e. root resorption) and accelerated tooth movement. The aim of the present study is to find out a standardized GCF collection, storage and total protein extraction method from apparently healthy gingival sites with orthodontics that is compatible with further high-throughput proteomics. Eighteen patient… Show more

“…The fact that weaker shear bond strength can be obtained by using a hydrophobic primer under contamination has been noted by previous in vitro studies [2,15,[27][28][29]. This hydrophobicity is attributed to its composition of 2, 2-bis[p-(20-hydroxy-30-methacryloxypropoxy) phenyl] propane/triethyleneglycol dimethacrylate (Bis-GMA/TEGDMA) with a ratio of 1/1 by weight.…”

“…Gingival crevicular fluid increases the number of bond failures [25]. Simple moisture exclusion using cotton rolls alone is unsuitable for dry-field techniques because of the 100% relative humidity [26,27]. Thus, saliva ejector and cotton roll isolation alone cannot maintain a 100% dryness of the enamel surface.…”

“…GCF samples were collected by the same trained clinician (C.B.) six weeks after the conclusion of the cause-related therapy (at baseline) and immediately before the periodontal surgery, following an intrasulcular absorption method [ 23 , 24 ]. Prior to GCF collection, the intervention site was isolated and dried with cotton rolls.…”
