Commentary Vav‐1 and T cells

Abstract: [I 23942]Vav-1 is a protein expressed predominantly in hematopoietic cells. There has been much work on the biochemistry and biology of this protein and finally some clarity as to the role of Vav-1 in immune cells has been achieved. An excellent paper on this topic was recently published by Ardouin et al. [1].The involvement of Vav-1 in lymphocyte transduction was first indicated by biochemistry -it was seen that this protein was tyrosine-phosphorylated in response to antigen-receptor ligation. Vav-1 is also t… Show more

“…Vav GEF activity is important for HIV replication, as dominant-negative Vav suppresses HIV replication when expressed in infected cells (Fackler et al, 1999). In the normal immune response, Vav plays a crucial role in TCR signaling events, including Ca 2+ flux and cytoskeletal reorganization (Cantrell, 2003). In turn, c-Cbl participates in turning off Vav activity through ubiquitination.…”

“…The importance of such Vav signalling is underlined by the finding that Vav1 −/− Vav2 −/− mice exhibit substantially reduced numbers of B cells which display impaired activation responses (reviewed in [5]). In addition to being a mediator of the actin polymerisation/cytoskeletal reorganisation which is required for capping of the antigen receptor, Rac1 enables optimal calcium signalling in lymphocytes [59,60] by contributing to the stimulation of PLC␥2 and PI 3K downstream of the BCR [16,21,35,59,[61][62][63][64]. Similarly, the stimulation of Vav/Rho downstream of the BCR can stimulate PI-4-phosphate-5-kinase, the lipid kinase that is responsible for maintaining the levels of PI-4,5-P 2 in cellular membranes and hence provides a constant supply of substrate for PLC␥ [60].…”

“…Although the best‐characterized GEFs that mediate the activation of Rac proteins in response to TCR stimulation are the Vav proteins (152, 153), additional classes of Rac activators exist in CD4 + T cells. Here we focus on Def6, a novel type of Rac activator that has emerged as a critical controller of T‐cell activation.…”
