Common variants in the CRP gene in relation to longevity and cause-specific mortality in older adults: The Cardiovascular Health Study

Abstract: Common polymorphisms in the CRP gene are associated with plasma CRP levels in populationbased studies, but associations with age-related events are uncertain. A previous study of CRP haplotypes in older adults was broadened to include longevity and cause-specific mortality (all-cause, non-cardiovascular (nonCV), and cardiovascular (CV)). Common haplotypes were inferred from four tagSNPs in 4512 whites and five tagSNPs in 812 blacks from the Cardiovascular Health Study, a longitudinal cohort of adults over age … Show more

“…However, in the same study, a positive association between þ 1846 AA homozygotes and risk of lung cancer was observed (OR 2.6; 95% CI 1.6 -4.4). In another study of elderly people, an association between CRP haplotype B, bearing the þ 1846 A-allele, and cancer-related death was found, but the cancer cases were not analysed separately according to site (Hindorff et al, 2008). This allele A is also part of the high-PSA-associated ACCCA haplotype in our study.…”

“…This is in contrast to a recent study by Leber et al 51 showing that Lactobacillus casei Shirota significantly increased hs-CRP in patients with metabolic syndrome. Elevated hs-CRP is independently associated with increased risk of CAD, 52 diabetes mellitus, 53 hypertension, 54 stroke 55 and mortality 56 and is closely associated with elevated fibrinogen. Fibrinogen as well as hs-CRP is regulated by IL-6 in hepatocytes.…”
