Communication Between Epithelial–Mesenchymal Plasticity and Cancer Stem Cells: New Insights Into Cancer Progression

Abstract: The epithelial–mesenchymal transition (EMT) is closely associated with the acquisition of aggressive traits by carcinoma cells and is considered responsible for metastasis, relapse, and chemoresistance. Molecular links between the EMT and cancer stem cells (CSCs) have indicated that EMT processes play important roles in the expression of CSC-like properties. It is generally thought that EMT-related transcription factors (EMT-TFs) need to be downregulated to confer an epithelial phenotype to mesenchymal cells a… Show more

“…Repressed production of ROS in high glucose treatment is in line with findings of previous investigation (4). ROS has a dual role to play in cancer biology; and its low level upkeeps the survival of cancer stem cells, which correlates with chemoresistance (44,45). Curcumin was able to augment the ROS production by doxorubicin even in the hyperglycemic condition.…”

“…The fact that CSCs can change phenotypes via different programs, such as dynamic epithelial/mesenchymal status, has led to the speculation that non-CSCs can transform back to CSCs ( 47 49 ). Therefore, CSC plasticity can be presumed as the cell state capable of being shaped by EMT, wherein this process can allow interconversion of CSCs and non-CSCs ( 50 53 ).…”

“…As a result, the less organized tumour structures create a local microenvironment conducive for increased cell mobility, which is thought to promote invasion and metastases [ 259 ]. Thus, EMT not only plays a key role in the generation, selection, and survival of tumour cells, it is simultaneously the product of enhanced mesenchymal or stromal-cell-like properties, and phenotypic heterogeneity [ 260 , 261 ].…”

“…Additional, very detailed studies were performed to further investigate the role of NOTCH4 in this context [ 315 ], also in other cancer entities. We strongly recommend two recent reviews, which cover the topic of epithelial–mesenchymal plasticity, TME and their contribution to developing CSC [ 260 , 316 ]. The alternatives for targeting the Notch signalling pathway in CSCs are summarized in a review by Venkatash et al [ 324 ].…”
