ComparativeIn VitroCytotoxicity Studies of177Lu-CHX-A″-DTPA-Trastuzumab and177Lu-CHX-A″-DTPA-F(ab′)2-Trastuzumab in HER2-Positive Cancer Cell Lines

Abstract: Background: Human epidermal growth factor receptor 2 (HER2) is found to be amplified in *15%-20% of breast cancers. In this study, the authors report the synthesis and comparative in vitro therapeutic efficacy of 177 Lu-CHX-A †-DTPA-trastuzumab and 177 Lu-CHX-A †-DTPA-F(ab¢) 2 -trastuzumab to determine their potential as theranostic agents for patients with breast cancer. Materials and Methods: Bivalent F(ab¢) 2 -trastuzumab was produced by enzymatic digestion of trastuzumab, conjugated with p-SCN-Bn-CHX-A †-D… Show more

“…Since their discovery in the late 1980s, the ITGB superfamily members are demonstrated to be involved in the regulation of cancer development and progression ( Cooper and Giancotti, 2019 ). The role of ITGBs in the tumorigenesis and pathogenesis of various cancer types has increasingly received attention ( Sharma et al, 2020 ; Fujita et al, 2021 ; Nurzat et al, 2021 ; Paindelli et al, 2021 ). Here, in this paper, we evaluate the utility of ITGBs as biomarkers in GC, and we utilize bioinformatics tools for investigating the underlying mechanisms by ITGB5 affected GC.…”

“…The concentrated antibody was added to 0.5 mL of immobilized pepsin (3000 U/mg), pre-equilibrated with digestion buffer. The preliminary experiments, which include the optimization conditions for enzymatic digestion, are reported elsewhere . Post preparation, the antibody fragments were purified using Amicon filtration devices (MWCO 30,000 Da) and stored in phosphate buffer saline (PBS, 20 mM, pH 7.4).…”

“…111 In, 125 I, 67 Ga) for radiation treatment of tumours (Aghevlian et al 2017 ; Ku et al 2019 ). Trastuzumab labeled with the β-particle emitter, 177 Lu was cytotoxic in vitro to HER2-positive human SK-BR-3 and MDA-MB-453 human BC cells and SK-OV-3 ovarian cancer cells (Sharma et al 2020 ). Treatment of athymic mice with intraperitoneal (i.p.)…”
