Comparative Mitogenomic Analysis Reveals Dynamics of Intron Within and Between Tricholoma Species and Phylogeny of Basidiomycota

Abstract: The genus of Tricholoma is a group of important ectomycorrhizal fungi. The overlapping of morphological characteristics often leads to the confusion of Tricholoma species classification. In this study, the mitogenomes of five Tricholoma species were sequenced based on the next-generation sequencing technology, including T. matsutake SCYJ1, T. bakamatsutake, T. terreum, T. flavovirens, and T. saponaceum. These five mitogenomes were all composed of circular DNA molecules, with sizes ranging from 49,480 to 103,09… Show more

“…Positive selection signals in fungal mitochondrial rps3 genes have been reported previously ( Lin et al, 2015 , 2017 ; Kang et al, 2017 ; Wang et al, 2020 ; Zhang et al, 2020 ; Huang et al, 2021 ; Wu et al, 2021 ). Together with rps3 , genes encoding ribosomal subunits with positive AT and GC skewness are identified in the mitogenomes of brown rot fungal pathogens ( Yildiz and Ozkilinc, 2021 ).…”

“…The intronic ORFs also share significant homologies with mitogenome intronic ORFs of unrelated fungal species (Supplementary Excel File 2), often outside the basidiomycetes, something commonly cited as evidence of introgression between diverse species. Similar intron dynamics were observed within the Agaricales (Huang et al, 2021).…”

“…For M. tenuissimus, the five isolates carrying a single cox1 intronic ORF show close similarity (>99% identity) while the 3 cox1 intronic ORFs of isolates 63 and 79 show close similarity (>99% identity) in a positionally dependent manner (Figure 6B). As seen in other comparisons, the genetic relationships among these intronic ORFs across species, in some cases, also show specificity as to position within specific core genes (Huang et al, 2021). For example, the terminal GIY-YIG containing intronic ORF in cox1 shows sequence similarity (>95%) across all species except M. tenuissimus (Figure 6A).…”

“… 2018 , 2019 ), but not for others (Huang et al. 2021 ). This study will help to better understand the phylogenetic status of Coprinellus and the mitogenome evolution in macrofungi, and will be of interest for further applied researches by selecting the most suitable species for biotechnological and nutritional interest.…”

“…The size of the complete mitogenome (64,450 bp) of C. micaceus was larger than that of Coprinopsis_cinerea, 42,448 bp (NW003307477.1, Stajich et al 2010). Recent researches indicated that the genome sizes of mitochondria in Agaricales varied significantly from about 40 kb (Paxillus involutus, MK993563, Li et al 2020) to more than 120 kb (Clavaria fumosa, MT114157, Wang et al 2020), sometimes that were near in a same genera or very closed genus (Yang et al 2016; Li et al 2018, 2019, but not for others (Huang et al 2021). This study will help to better understand the phylogenetic status of Coprinellus and the mitogenome evolution in macrofungi, and will be of interest for further applied researches by selecting the most suitable species for biotechnological and nutritional interest.…”
