Comparative proteome analysis of Helicobacter pylori clinical strains by two-dimensional gel electrophoresis

Abstract: Abstract:Objective: To investigate the pathogenic properties of Helicobacter pylori by comparing the proteome map of H. pylori clinical strains. Methods: Two wild-type H. pylori strains, YN8 (isolated from biopsy tissue of a gastric cancer patient) and YN14 (isolated from biopsy tissue of a gastritis and duodenal ulcer patient), were used. Proteomic analysis, using a pH range of 3-10 and 5-8, was performed. The individual proteins were identified by quadrupole time-of-flight (Q-TOF) mass spectrometer and prote… Show more

“…In a previous study, we investigated the comparative proteome of clinical H. pylori strains by two-dimensional gel electrophoresis. We reported differential proteins of H. pylori strains isolated from gastritis and gastric cancer, including antioxidants (Zhang et al, 2011). In this study, we investigated the expression of three essential antioxidants of H. pylori in clinical isolates to show the relationship between the expression of these antioxidants and different gastric diseases.…”

“…strains within each cluster. Given the complexity and high costs of the technology, proteomic studies are, in general, limited in respect to sample size (Smith et al 2007;Franco et al 2009;Zhang et al 2009Zhang et al , 2011. Our study is not an exception, and therefore, to achieve our goal, we sought to minimize this issue by selecting for the analysis the 2DE maps [previously obtained in (Vitoriano et al 2011a)] of the group of 11 epidemiologically and genetically unlinked H. pylori strains used in the restricted dendrogram ( Fig.…”

“…For patients with positive screening results, general practitioners will conduct a comprehensive assessment based on the patient's symptoms, signs, and possible risk factors. When necessary, they will recommend more accurate diagnostic methods such as gastroscopy and biopsy [17] . However, due to the limited popularity of gastroscopy in primary care institutions, general practitioners often need to rely on their clinical experience and judgment, as well as collaboration with higher-level medical institutions, to provide optimal treatment recommendations for patients.…”
