Comparative Study of the Efficacy of Olmesartan/Amlodipine vs. Perindopril/Amlodipine in Peripheral and Central Blood Pressure Parameters After Missed Dose in Type 2 Diabetes

Abstract: The OLM/AML combination is safe, well tolerated, and not inferior to the combination of PER/AML in lowering CBP and CBP-derived parameters in diabetic patients. OLM/AML provides longer-lasting efficacy in terms of CBP reduction compared to PER/AML.

“…In the case of oral contraceptives, this was done to define the interval between a last-taken pill before the ovulationinducing surge of pituitary gonadotrophins appeared [21]. Similar studies have been performed with antihypertensive drugs [22][23][24][25][26][27][28][29][30] which indicate, for instance, amlodipine to be more forgiving than either enalapril or diltiazem, and to be more forgiving when combined with olmesartan than with perindopril [31]. For warfarin, pharmacokineticpharmacodynamic models have been proposed to inform labelling recommendations [32], and population pharmacokinetic models have been used to simulate missed doses of [33].…”

“…Of note, the incidence of drug-related adverse events was largely independent of dose titration and was, in fact, lower when the drug was used in fixed-dose combination therapies with HCTZ, AML or both [ 46 49 ]. In addition, a recent randomised, double-blind, controlled clinical study demonstrated the non-inferiority of the fixed-dose combination of OLM and AML to the combination of perindopril and AML in reducing diastolic BP after 24 weeks of treatment and after a missed dose (− 11.7 and − 10.5 mmHg, respectively) [ 50 , 51 ].…”
