ABSTRACT. This study evaluated the mutagenicity and antimutagenicity of inulin in a chromosomal aberration assay in cultures of the meristematic cells of Allium cepa. The treatments evaluated were as follows: negative control -seed germination in distilled water; positive control -aqueous solution of methyl methanesulfonate (10 μg/mL MMS); mutagenicity -aqueous solutions of inulin (0.015, 0.15, and 1.50 μg/mL); and antimutagenicity -associations between MMS and the different inulin concentrations. The antimutagenicity protocols established were pretreatment, simultaneous simple, simultaneous with pre-incubation, and post-treatment. The damage reduction percentage (DR%) was 43.56, 27.77, and 55.92% for the pre-treatment; -31.11, 18.51, and 7.03% for the simultaneous simple; 30.43, 19.12, and 21.11% for the simultaneous with pre-incubation; and 64.07, 42.96, and 53.70% for the post-treatment. The results indicated that the most effective treatment for inhibiting damages caused by MMS was the post-treatment, which was followed by the pretreatment, suggesting activity by bioantimutagenesis and desmutagenesis.The Allium cepa assay was demonstrated to be a good screening test for this type of activity because it is easy to perform, has a low cost, and shows DR% that is comparable to that reported studies that evaluated the prevention of DNA damage in mammals by inulin.

Background/Objective: Carboxylic acids (CAs), especially butyric acid, have been suggested to counteract colonic diseases, such as ulcerative colitis and colon cancer. Colonic formation of CAs can be influenced by the diet, but the concentrations and pattern formed need to be evaluated for different food products in humans. To elucidate how the colonic concentration of CAs in healthy subjects is influenced by dietary supplementation with oat bran, and whether the concentration varies over time and during consecutive days. Subjects/Methods: Twenty-five healthy subjects (age 2471.3) were recruited to the study. The subjects were given 40 g b-glucan enriched oat bran per day, corresponding to 20 g dietary fibre, in 4 slices of bread. CAs were analysed in faeces during three consecutive days after 0, 4, 8 and 12 weeks on this diet. Results: The concentration of acetic, propionic, butyric, isobutyric and isovaleric acid was higher (Po0.05-0.001) after 8 weeks on the oat bran diet as compared with values at entry, whereas that of lactic acid was lower (Po0.05). After 12 weeks, the concentrations of acetic, propionic and isobutyric acid were still higher and that of lactic acid lower. The variation between individuals was considerable, whereas in the same individuals there was little variation. Conclusions: Oat bran increased the faecal concentration of CAs after 8 weeks, indicating an increased concentration also in the distal colon. The concentration of all main acids increased, except for lactic acid, which decreased. Oat bran may therefore have a preventive potential adjunct to colonic diseases.

In vivo fermentation of Ulva lactuca was studied in previously germ‐free rats inoculated with human flora obtained from non‐, low‐ and high‐methane producers (groups NMP, LMP and HMP, respectively), in comparison to germ‐free rats. Rats were fed either a control diet or a diet containing 4% of dried Ulva lactuca. Production of metabolites varied according to the flora and diet. Ulva lactuca induced a specific high production of methane in the HMP group. With the three human flora, Ulva diet induced a similar increase in caecal pH. In the NMP and HMP groups, this increase was associated with a fall of lactic acid caecal concentration. In the LMP group it was related to a decrease in the concentration of short‐chain fatty acids. Ulva lactuca appeared to be able to regulate the β‐glucuronidase and β‐glucosidase activities, reducing the relatively high levels observed in groups NMP and HMP and increasing the low levels obtained in the LMP group. Results show that, although it was poorly fermented, Ulva induced significant effects on the gut microflora metabolism. The methanogenic status of the human donor appeared to be an important factor. © 1998 SCI.
