Comparative study of the physiological roles of three peroxidases (NADH peroxidase, Alkyl hydroperoxide reductase and Thiol peroxidase) in oxidative stress response, survival inside macrophages and virulence of Enterococcus faecalis

Abstract: SummaryThe opportunistic pathogen Enterococcus faecalis is well equipped with peroxidatic activities. It harbours three loci encoding a NADH peroxidase, an alkyl hydroperoxide reductase and a protein (EF2932) belonging to the AhpC/TSA family. We present results demonstrating that ef2932 does encode a thiol peroxidase (Tpx) and show that it is part of the regulon of the hydrogen peroxide regulator HypR. Characterization of unmarked deletion mutants showed that all three peroxidases are important for the defence… Show more

“…When required, erythromycin (150 mg ml 21 ) was added to E. faecalis cultures. Cultures of E. faecalis strains were also performed on CCM17 MOPS medium with glycerol (0.5 %, v/v) as the sole carbohydrate (CCM17-gly), as described by La Carbona et al (2007). Different pH conditions (4.0-9.0) were tested using microplates with CCM17-gly medium.…”

“…The most conserved oxidative resistance mechanism in LAB results from the coupling of NADH oxidase and NADH peroxidase (184 (247). Multiple enzymes with peroxidase activity have been described for LAB, such as AhpCF, Npr, and Tpx of E. faecalis and AhpCF and Tpx of S. mutans (248).…”

“…An alternative strategy to counteract superoxide anion radicals is by the production of high levels of intracellular glutathione (252). Various enzymes that are known to counter oxidative stress, namely, MsrAB, AhpCF, Tpx, Npr, KatA, Sod, and HypR, have been characterized for E. faecalis (94,248,(253)(254)(255).…”

“…Other reports indicate that Tpx is expressed extracellularly by Mycobacterium tuberculosis (84), Actinobacillus actinomycetemcomitans (38), and Bacillus anthracis (2) and periplasmically in E. coli (16), despite the apparent absence of a signal sequence. Interestingly, Tpx also appears to play a role in protecting Enterococcus faecalis against phagocytosis (62). If analogous to H. pylori, this may explain, in part, H. pylori's resistance to phagocytic digestion.…”
