Measurement of thyroid hormones and cortisol in horses withan automated immunoassay analyzer Abstract: Even though thyroid gland diseases are unusual in horses, it is important to have a reference interval for thyroid hormones in this species at every veterinary endocrine laboratory. In addition, cortisol and thyroid hormone measurement is useful in horses, not only for disease diagnosis, but as a tool for monitoring and researching performance and metabolic rate.The objectives of this study included (i) to test the capacity of the automated analyzer AIA 360® (TOSOH Bioscience) to measure thyroid hormones and cortisol in equine serum in a reliable way, and (ii) to establish a reference interval for thyroid hormones in Costa Rican horses using the AIA-360®.A total of 68 healthy and treatment free horses (31 males and 37 females) were sampled. Horses were grouped into three different categories according to their ages. Serum concentrations of total thyroxine (TT4), free thyroxine (FT4), and cortisol were quantified with the AIA-360®. Statistic relationships of hormonal values, gender and age were determined using Kruskal-Wallis, Dunn and Wilcoxon (U Man-Whitney) tests. P <0.05 was considered significant.Selected reference intervals were: TT4 14.16 -46.33 nmol/L, FT4 3.60 -16.09 pmol/L and cortisol 23.45 -166.64 nmol/L. No statistically significant relationship was found between gender, age, and hormone values. Besides, significant relationships (p<0.05) were found between hormone levels and some environmental parameters. Animals living at higher altitudes showed higher cortisol levels, while cortisol decreases as the environmental temperature rises. Moreover, the FT4 levels are higher as precipitation increases. Finally, TT4 seems to be lower as day light length increases; however, no statistical relationship was found (p<0.30). Our results show that the use of the automated analyzer AIA-360® is suitable to measure thyroid hormones and cortisol, and that the values found are comparable with other studies and analysis techniques.Keywords: horses, thyroid hormones, cortisol, reference interval, AIA-360®Resumen: A pesar de que las enfermedades tiroideas son poco comunes en equinos, es importante contar con un intervalo de referencia para hormonas tiroideas en cualquier laboratorio de endocrinología veterinaria. We evaluated the relationship between thyroid hypofunction and insulin resistance among obese female patients visiting to endocrinology outpatient clinics (OPD) done at Sparsh Endocrinology & Diabetic Center. In this cross-sectional study, a total of fifty female subjects with age group of 30-60 years, whose body mass index was above 30 kg/m2 were studied for 16 months between July 2013 to November 2014 who were attending our endocrinology OPD were studied. Subjects with known disease or disorder of endocrine systems or who were receiving medications like, thyroid hormones preparations, Lithium, Amiodarone or corticosteroids were excluded. Blood samples were assed for T3, T4, TSH, FBS, Fasting Insulin and HOMA IR was calculated. A total of fifty subjects participated in the study. The mean age of the patients was 31.4 ± 7.32 years. Among these subjects TSH analysis revealed that 26% were having subclinical hypothyroidism (SCH). We found that 46% of these SCH subjects are having insulin resistance while 24% of patients with normal thyroid function are having insulin resistance as estimated by HOMA IR. P-value=0.6 and the odds ratio is 0.667, with a 95% confidence interval extending from 0.19 to 2.09. The observed difference in insulin resistance was not statistically between the subjects with SCH and normal obese subjects. In our study we observed that insulin resistance co exists with thyroid-hypo function in 46% of obese subjects. Caution has to be executed in interpretation of results as our sample is small.

Electrogenerated chemiluminescence (electrochemiluminescence, ECL) generates species at electrode surfaces, which undergoes electron-transfer reactions and forms excited states to emit light. It has become a very powerful analytical technique and has been widely used in such as clinical testing, biowarfare agent detection, and pharmaceutical analysis. This review focuses on the current trends of molecular recognition-based biosensing methods for pharmaceutical analysis since 2010. It introduces a background of ECL and presents the recent ECL developments in ECL immunoassay (ECLIA), immunosensors, enzyme-based biosensors, aptamer-based biosensors, and molecularly imprinted polymers (MIP)-based sensors. At last, the future perspective for these analytical methods is briefly discussed.
