Comparison of Staphylococcus Phage K with Close Phage Relatives Commonly Employed in Phage Therapeutics

Abstract: The increase in antibiotic resistance in pathogenic bacteria is a public health danger requiring alternative treatment options, and this has led to renewed interest in phage therapy. In this respect, we describe the distinct host ranges of Staphylococcus phage K, and two other K-like phages against 23 isolates, including 21 methicillin-resistant S. aureus (MRSA) representative sequence types representing the Irish National MRSA Reference Laboratory collection. The two K-like phages were isolated from the Fersi… Show more

“…Most staphylophages detected in commercial preparations belong to the Myoviridae family, namely to the Kayvirus 4 , 24 . The Myoviridae phages are associated with the lytic life cycle and characterized by the high efficiency of the bacterial lysis.…”

“…Host range determination of individual bacteriophages and the ability of therapeutic phage cocktails to produce plaques were performed by the plaque assay as previously described with some modifications 4 , 5 , 24 . Five microliters of phage lysate at a titer of 10 6 PFU/ml or five microliters of the commercial therapeutic bacteriophage cocktail was spotted on indicator plates, which contained 100 µl of late log phase cultures of investigated strains (10 6 CFU) and 5 ml of top LB agar (0.6% w/v).…”

“…Isopycnic centrifugation through CsCl gradients was performed as previously described, with modifications (Ajuebor et al, 2018). A high titer phage lysate was precipitated using polyethylene glycol (15% w/v PEG8000, 0.5M NaCl) at 4 • C overnight and centrifuged at 12000 × g for 20 min at 4 • C. Thereafter, the pellet was resuspended in SM buffer.…”

“…Even though the Stab phages had distinct host ranges, with Stab20 and Stab21 having a wide and Stab22 and Stab23 a narrow host range, their genomes proved to be closely related to other phages of therapeutic and biocontrol significance such as phage Sb-1 [31,57]. Proteomic analysis depicted uniformity among some of their structural proteins with the Staphylococcus phage K and other members of Kayvirus genus [58]. These Stab phages were stable at temperatures below 40 • C, between pH 5.4-9.4, and could resist U.V energy up to 25 µJ/cm 2 .…”
