Compartments and Cell Flows Within the Mouse Haemopoietic System

Abstract: Part-body irradiated CBA mice were injected with CBA-T6 bone marrow. In this way a predominantly donor population was established in the femora while the marrow of the humeri remained largely (average 94 %) of host origin. In animals examined cytologically up to 2 years later, no tendency was observed for the proportion of donor cells in the humeri to increase. Splenectomy had no effect on this. When femoral bone marrow from the experimental mice was injected into lethally (whole-body) irradiated recipients, c… Show more

“…Creation of localized BMT was evident from the remarkable increase in femur cellularity in the perfused limb, and the extremely low incidence of PKH + cells in peripheral blood and contralateral femurs. Considering that IL-BMT created localized engraftment and systemic distribution of few cells, it is likely that BMC were mobilized from the injected limb at a later time [18]. Secondary systemic redistribution was apparent in the levels of donor chimerism at 30 days (24%-76%) that exceeded by far the contribution of one femur and tibia (7%-9%) to the hemopoietic space in the mouse [19].…”

“…Busulfan is a cytoreductive agent that selectively affects slow-cycling and quiescent BMC, predominantly primitive stem cells in G 0 [24]. It is likely that the systemic preconditioning procedure used in this study promoted the rates of secondary systemic seeding of locally transplanted BMC [18,25].…”

“…In our previous experiments, with either fetal liver (5) or adult bone marrow (7) grafts, progressive replacement of donor-type RBC was invariably accompanied by coordinate re- Hematopoiesis in adult irradiated or W-anemic mice has previously been found to be restored from a small number of stem cells in donor bone marrow, as estimated from unique chromosomal markers (2,(16)(17)(18) or clinical cure of the host's anemia by limiting dilutions of cells (19). Results in the present experiment support the conclusion that only a single stem cell from the small donor inoculum of fetal liver has Fig.…”

“…The levels of donor chimerism at 3-4 weeks exceeded the contribution of one femur and tibia (8%-9%) to the total hematopoietic space in the mouse [32], which would be expected if engraftment remained localized over a long period. Redistribution of cells in the host bones via a hematogenic route and secondary engraftment [33] is a process that proceeds at a relatively slow pace, and its rate is proportional to the number of cells infused [34]. At 4 weeks, the noninjected femurs of myeloablated hosts contained ~15% of the donor cells found in femurs of injected limbs, and the fraction of stem cells (defined as Sca-1 + c-kit + lineage -) was 2.5-fold lower.…”
