Complete Genome Sequence of Staphylococcus aureus M013, a pvl-Positive, ST59-SCCmec Type V Strain Isolated in Taiwan

Abstract: We report the complete genome sequence of M013, a representative strain of a pvl-positive, sequence type 59-staphylococcal cassette chromosome mec type V (ST59-SCCmec type V) community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) clone in Taiwan. Comparison of M013 with the genomes of two CA-MRSA strains in the United States revealed major differences in the regions covering several genomic islands and prophages.

“…Lastly, the complete genome sequence of an ST59 CA-MRSA isolate, a lineage predominant in Taiwan, was reported recently (Huang et al, 2012a, 2012b). Since the original identification of ST59, these isolates have frequently been associated with multi-drug resistance.…”

“…Since the original identification of ST59, these isolates have frequently been associated with multi-drug resistance. Compared to the USA300 strain FPR3757 and the USA4000 strain MW2, most of the open reading frames in the ST59 strain M013 were highly conserved (Huang et al 2012a). However, genomic islands SaPI5 and νSa3 as well as prophages ϕSa3usa and ϕSa3 are absent in M013 (Huang et al, 2012a, 2012b).…”

“…However, an ST59 community-associated MRSA clinical Taiwanese strain, M013 (43), which shares three cmk synonymous and cmk(S203T) mutations with the two ST45 strains, is susceptible to vancomycin (MIC, Յ0.5 mg/liter) (T. L. Lauderdale, personal communication). Therefore, it is not clear if cmk(S203T) is directly associated with raised vancomycin resistance or not.…”

“…To broaden our survey, we analyzed cell-free culture extracts of 12 additional S. aureus strains or clinical isolates ( Table S1) . N-AgrD peptides were produced by three USA300 lineage MRSA, two ST59 MRSA (Huang et al, 2012), and clonal colony ST45 of USA600 MRSA lineage ( Figure 1C ). In successive MS experiments, peptide sequences most abundantly mapped to AgrD (McIlwain et al, 2012), herein designated (i) N-AgrD F20, (ii) N-AgrD F24, and (iii) N-AgrD D20, were synthesized and isolated at >95% purity for functional assessment.…”
