Complex-specific immunoglobulin M antibody patterns in humans infected with alphaviruses

Abstract: Sera from humans with serologically confirmed eastern equine encephalitis, western equine encephalitis, Pogosta (Ockelbo), Mayaro, Ross River, and chikungunya virus infections were tested by immunoglobulin M (IgM) antibody capture enzyme immunoassay. Diagnostically useful IgM antibody titers were detected, and selected sera with high IgM antibody titers were tested for IgM antibody with nine heterologous alphaviruses. The results provide evidence for the complex specificity of IgM antibody and indicate the use… Show more

“…This is reminiscent of responses to some other groups of antigenically related viruses (e.g. alphaviruses) where cross-reactive IgM responses have been documented [Calisher et al, 1986]. The viral antigens in the CMV IgM ELISA assay may be different to those relevant in the ACIF assay to detect rising antibody titres to CMV.…”

“…In a broader context, analysis of IgM response in humans with alphaviruses infection has revealed the potential usefulness of IgM as a diagnostic tool especially during endemics. 53 Indeed, IgM-based ELISA has been developed for CHIKV serodiagnosis of adult patients infected with La Reunion strain. 54 At present, there are no prior studies done on understanding the use of CHIKV-specific IgM as an immunotherapy in patients and animal models.…”

“…There is too little information relating to IgM antibody responses in anamnestic a1phavirus infections in humans to determine whether these three apparent dual infections with Ross River and Barmah Forest viruses could have been due to a primary infection with one virus and an anamnestic response to antigens shared between this virus and one causing an earlier, perhaps subclinical, infection. However the low levels of cross-reactivity between alphavirus IgM antibodies makes this unlikely.,, 22 As well as collecting the data necessary to complete a comprehensive picture of clinical Barmah Forest virus disease, tile interval from infection to the onset of symtoms needs to be determined, as does the duration of viraemia. A knowledge of the duration of viraemia and of the incidence in the community (estimated to be 0.23% of the population per annum in Queensland) may indicate whether the continuous transmission of Barmah Forest virus requires a non-human host or whether human-mosquito-human cycles are liIely to be self-sustaining.…”
