Comprehensive proteomics and functional annotation of mouse brown adipose tissue

Abstract: Knowledge about the mouse brown adipose tissue (BAT) proteome can provide a deeper understanding of the function of mammalian BAT. Herein, a comprehensive analysis of interscapular BAT from C57BL/6J female mice was conducted by 2DLC and high-resolution mass spectrometry to construct a comprehensive proteome dataset of mouse BAT proteins. A total of 4949 nonredundant proteins were identified, and 4495 were quantified using the iBAQ method. According to the iBAQ values, the BAT proteome was divided into high-, m… Show more

“…Of note, there is a lack of consensus in the published data concerning the gene and protein expression of CD248 in brown adipose tissue. Here, in the mouse, we detected similar levels of mRNA in white (gonadal, inguinal, and peri-renal) depots as compared to scapular brown fat, confirming the findings of Roh et 2020) did not detect it in murine BAT [25,26]. The phenotypes we have detected in the Cd248 -/mouse are all seen in the WAT compartment, where Cd248 expression is not disputed, however we highlight this uncertainty to the reader for completeness and consideration when interpreting the findings.…”

“…Generally, Hb is conserved in vertebrates and is often used as a molecular clock [7]. In bats, the high hematocrit and Hb levels enable them to obtain enough oxygen [23]. In the present proteomics study, Hemoglobin subunit is identi ed in the Indian false vampire bat, Megaderma lyra.…”

“…Last but not least, although the plasma estradiol level was significantly decreased in the Ovx group, the direct causal relationship with estrogen deficiency cannot be proven due to no intervention group with estradiol to access the reversibility of these proteomic changes. Moreover, the BAT proteome might be influenced in different environments such as thermoneutrality, cold exposure, or a high-fat diet 12 , 20 , 70 . Further studies investigating the BAT proteomic change under different temperatures or diets as well as the reversibility under estrogen replacement might be important to provide a deeper understanding of the relationship between BAT, estrogen, and environmental factors.…”

“…Proteomic analysis is a comprehensive protein study that provides information on changes in protein expression along with their interacting networks and identifies biomarkers related to pathogenic processes 19 . Proteomic techniques have demonstrated their usefulness in investigating the biological function of BAT 20 , 21 . To our knowledge, no comparative study on the proteomic changes of BAT in ovariectomized rats has been conducted.…”
