Computel: Computation of Mean Telomere Length from Whole-Genome Next-Generation Sequencing Data

Abstract: Telomeres are the ends of eukaryotic chromosomes, consisting of consecutive short repeats that protect chromosome ends from degradation. Telomeres shorten with each cell division, leading to replicative cell senescence. Deregulation of telomere length homeostasis is associated with the development of various age-related diseases and cancers. A number of experimental techniques exist for telomere length measurement; however, until recently, the absence of tools for extracting telomere lengths from high-throughp… Show more

“…Whole genome sequencing (WGS) -based TL measurement techniques (34) are new promising tools that use WGS data to calculate TL ( Figure 1). Some studies have already calculated TL with bioinformatics tools such as Computel, Motif_counter, TelSeq and qMotif (34)(35)(36)(37)(38)(39)(40). These methods had a high level of concordance with q-PCR measurements (34).…”

“…Most studies use a qPCR based method, where differences in the standard used can affect study results and make comparisons between studies and laboratories difficult. Alternate approaches comprise the measurement of absolute telomere length by pulsed field gel electrophoresis [292], which is more labor-intensive and therefore less readily applicable to large studies, and computation [293].…”

“…Telomere is a nucleoprotein complex residing in the termini of chromosomes with a repetitive DNA sequence of d (TTAGGG) [6,7] , which protects the chromosome ends from the recognition by the DNA-damage repair machinery and the enzymatic degradation. Its characteristic is that the length of the telomeric 3'-single strand is longer than that of the 5'-strand.…”
