Conjugated Polymer Nanoparticles with Appended Photo‐Responsive Units for Controlled Drug Delivery, Release, and Imaging

Abstract: Carriers that can afford tunable physical and structural changes are envisioned to address critical issues in controlled drug delivery applications. Herein, photo-responsive conjugated polymer nanoparticles (CPNs) functionalized with donor-acceptor Stenhouse adduct (DASA) and folic acid units for controlled drug delivery and imaging are reported. Upon visible-light (λ=550 nm) irradiation, CPNs simultaneously undergo structure, color, and polarity changes that release encapsulated drugs into the cells. The back… Show more

“…Additionally, the ROS generation efficiency of the nanosensitizer under X-ray irradiation was tested using the singlet oxygen sensor green (SOSG) agent. 30 As shown in Fig. S15 in the ESI, † the uorescence intensities increased with the dose in the solution containing MnTCPP-Hf-FA or TCPP-Hf-FA, suggesting that the assynthesized MOF NPs could produce ROS under X-ray irradiation.…”

“…Contrary to other switching molecules, such as azobenzenes, DASA reaction only requires Vis light and not UV. An example of this reaction can be found in the use of nanoparticles to encapsulate and release doxorrubicin (DOX) and camptothecin (CPT) for drug delivery . These nanoparticles were formed by self‐assembly of a two block PPV copolymer, one block functionalized with hydrophilic folic acid and the other with hydrophobic DASA molecules.…”

“…Over the past few years, semiconducting polymer dots (Pdots) have emerged as a new class of fluorescent probes for cellular imaging, sensors, in vivo imaging, and optical theranostics . Most reported Pdots with high fluorescence quantum yield (Φ f ) were based on a blue‐light‐excitable polymer host, such as poly(9,9‐dioctyl‐2,7‐fluorene) (PFO, 405 nm laser) and poly(fluorene‐alt‐benzothiadiazole) (PFBT, 488 nm laser) .…”
