Connecting nuclear architecture and genomic function

Abstract: Research in our laboratory employs a dual approach to correlating genomic function and regulation with nuclear architecture. In one series of projects we are identifying, cloning, and studying the molecular, genetic, and functional properties of the nuclear matrix proteins which comprise the three-dimensional nuclear architecture. In a second series, we are using molecular labeling, microscopic, and computer imaging approaches to visualize in three dimensions sites of genomic organization and function within t… Show more

“…There is increasing acceptance that components of nuclear architecture (chromatin, nuclear scaffold) are functionally linked to the organization and sorting of regulatory information in a manner that permits selective utilization (Berezney et al, 1996;Zeng et al, 1997Zeng et al, , 1998Lamond and Earnshaw, 1998;Ma et al, 1998Ma et al, , 1999McNeil et al, 1998McNeil et al, , 1999Choi et al, 2001;DeFranco, 2002;Gasser, 2002).…”

“…They were placed on ice, washed with Tris-bu er (50 mM Tris-HCl pH 7.5, 3 mM MgCl 2 , 0.5 mM PMSF), incubated 10 min in ice cold Tris-bu er with 0.5% Triton X-100, 0.5 mM CuCl 2 , then incubated at 378C for 30 min with Tris bu er containing DNAse I (50 mg/ml) and RNAse-A (20 mg/ ml) according to Berezney et al (1996). After washes, cells were incubated in 2 M NaCl in Tris bu er for 30 min on ice and ®xed in 2% PFA in PBS.…”

“…The nuclear matrix is a network of proteins throughout the inside of nucleus, which provides a structural framework for maintaining spatial order within nucleus and for proper nuclear functions , such as DNA replication and repair, gene transcription, RNA splicing and transport (Berezney, 2002;Berezney et al, 1996;Hancock, 2000). It is tempting to speculate that nuclear actin acts as a component of intranuclear filament network (or nucleoskeleton) that is analogous to cytoskeleton.…”
