Consecutive Use of Two Multiplex PCR-Based Assays for Simultaneous Identification and Determination of Capsular Status of Nine Common Neisseria meningitidis Serogroups Associated with Invasive Disease

Abstract: We developed two Neisseria meningitidis multiplex PCR assays to be used consecutively that allow determination of the serogroup and capsular status of serogroup A, B, C, 29E, W135, X, and Y cnl-3/cnl-1-likecontaining N. meningitidis isolates by direct analysis of the amplicon size. These assays offer a rapid and simple method of serogrouping N. meningitidis.

“…For example, in the European Union, 25% of cases are confirmed by PCR alone, and in the UK and Ireland, this rises to ~50% [20]; while in the United States, the CDC only recently included PCR as confirmatory in its case definition [6]. There is as yet no international consensus on their use, although PCR methods incorporating a number of targets have been developed (such as ctrA, porA, crgA , 16S rDNA, sodC and cnl ) [2125]. …”

“…Characterisation of N. meningitidis isolates using serotyping, genogrouping, porA/fetA finetyping, multilocus restriction typing (MLRT) and MLST was also undertaken [13][14][15][16][17][18]. Antimicrobial susceptibility testing was performed on isolates as above.…”
