Aim: The production of healthy cuttings from a local cassava cultivar for cassava mosaic control. Study design: The study was carried out at the In Vitro Culture Laboratory using the techniques of thermotherapy and culture of tissues and explants in a specific medium. Place and duration: The study was carried out at the Laboratory of Biological and Agronomic Sciences for Development at the University of Bangui, Central African Republic from December 2017 to June 2018. Methodology: A variety of cassava called six-month very susceptible to mosaic was used for this work. The cuttings used were infected by Cassava Mosaic Begomoviruses (CMBs) with high severity. It were subcultured in a room under the heat of 37 ° C to 40 ° C for two weeks. Explants and meristems were taken from the stems and the apices, respectively. These collected materials were treated and seeded on appropriate culture media. After the plants produced in vitro were acclimatized and the leaves were removed to check their phytosanitary state by the PCR technique. Results: The rate of emergence of the acclimatized plants and the expression of the disease on the microplants were evaluated. The results show that 75% of the weaned vitro plants recovered under acclimatization. In addition, the acclimatized plants left growing in the greenhouse for four months remained asymptomatic. Molecular analysis by PCR showed that begomoviruses were not detected on meristem samples unlike samples from stem fragments. Conclusion: The combination of thermotherapy technique associated with the culture of meristems constitutes an effective means useful for the sanitation of infected plant material.

Cassava In vitro regeneration is essentially influenced by exogen factors as well as explants nature. This work aimed to evaluate explants type and positions influence, phytohormone on cassava plantlets regeneration in modified MS media. The apex and nodes of the position 3, 5, and 7 were initiated on Murashige and Skoog (MS) based media supplemented with Gamborg B5 vitamins and growth regulators such as naphthalene acetic acid (NAA), 6-benzylaminopurine (BAP) and zeatin (ZEAT) in different concentrations. The results indicated a significant difference (p=0.0001) among explants sprouted regarding the nodes position and the cultivar genotype during the time of culture. The highest rate of regenerated plantlets was found with nodes of position 5 (95.33%) followed respectively by nodes of the position 7 (93.33%), nodes of the position 3 (89.33%), and the apex (76%). Regarding the growth regulators, NAA (0.5 mg/l) + BAP (0.5 mg/l) combination was favorable for height growth (26.2cm), and leaves development (7.84 cm) in most of cultivars, whereas NAA (0.5 mg/l) + ZEAT (0.5 mg/l) combination was more favorable to rhizogenesis in overall cultivars. Among the cultivars, the best performance in stem height (26.2 cm), in leaf length (7.84 cm), and roots number (4.64) were observed with INA 1er cultivar. The cultivars RB89509, BEN 86052, and DOKU were less grown on MS supplemented with 0.5 mg/l of BAP or ZEAT. These findings could contribute of using nodes of the positions 5, 7 as potential explants and Zeatin for cassava plantlets rhizogenesis.

Securidaca longepedunculata Fresen. is an overexploited forest species in the Lubumbashi region (south-eastern DR Congo), as its roots are highly valued in traditional medicine. Conventional propagation of this species is affected by seed dormancy and a high mortality rate during early seedling development. To improve on existing methods, we developed an in vitro seed germination protocol. After observing the germination rates, the effects of different doses (0.5, 1, 1.5, and 2 mg/L) of cytokinins (6-benzylaminopurine, kinetin, and meta-topolin) on S. longepedunculata seedling development were compared. Our results showed that soaking for 10 min in NaOCl (10%) followed by 5 min in ethanol (70%) effectively reduced the death rate of seeds while increasing the germination rate to almost 77%. The addition of cytokinins improved plantlet growth: a 12.2× increase in the number of plantlets was obtained with 1.5 mg/L meta-topolin, while only a single stem was obtained from the control. The effects of different auxin types on rhizogenesis did not differ significantly. The best recovery and rooting were noted with microcuttings from the basal parts of S. longepedunculata plantlets. Finally, the seedlings produced survived during the acclimatisation phase regardless of the type of substrate used. The established protocol provides a means for large-scale production of S. longepedunculata plantlets for the restoration of degraded landscapes and agroforestry.
