Cancer therapy has made use of plasmonic nanostructures for a long time. Platinum, gold, and silver are a few metals that can exhibit the SPR (surface plasmon resonance) phenomenon. In comparison to polymers and other metal nanoparticles, the SPR effect makes these nanostructures great PT (photothermal) agents for cancer treatment. In this study, we have synthesized hexagonally shaped gold nanorings (AuNRgs) with a 40 ± 10 nm diameter, which are applied as photothermal agents for treating melanoma. Results demonstrated that synthesized hexagon AuNRgs possess a great killing capacity against the B16 melanoma cell lines. Hexagon AuNRgs not only destroy cells via the PT effect but also enhance reactive oxygen species (ROS) generation to a great extent in the absence of any synthetic photosensitizer or near-infrared (NIR) dye. Hexagon AuNRgs showed an alteration in mitochondrial membrane potential with a significant green shift of the JC-1 dye. It was also observed that hexagon AuNRgs disrupted the blood vessels in the in-ovo CAM (chick chorioallantoic membrane) model. These findings support the utilization of hexagon AuNRgs as an anticancer agent against B16 melanoma.

Cancer is a complex disease that displays physiomorphological transformation in different surrounding microenvironments. Therefore, the single treatment modalities are relatively less effective, and their efficiency varies with tumor cell physiology, leading to the development of tumor resistance. Combinatorial therapeutic approaches, such as chemo-photothermal therapy, are promising for efficiently mitigating tumor progression irrespective of cancer physiology. Nanotechnology has played a significant role in this regard. Therefore, the present study reports the synthesis of poly(acrylic acid)-tetraethylene glycol (PAA-TEG)-coated BaSO4 nanoparticles (NPs) with enhanced solubility, dispersibility, and X-ray attenuation. Next, nanocomposites (NCs) are synthesized by loading BaSO4 NPs with the therapeutic drug triiodobenzoic acid (Tiba) and the photosensitizer IR780 using a lipid coating. These fabricated NCs are analyzed for dual-modal imaging (fluorescence and X-ray-based imaging) properties and chemo-phototherapeutic ability against two-dimensional (2D) and three-dimensional (3D) cultures of A549 cells. Furthermore, A549 cells are morphologically and physiologically aggravated into potent malignant cells using tobacco leaf extract (TE), and the variation in the therapeutic effect of NCs compared to cisplatin is determined. The synthesized NCs display enhanced encapsulation and excellent synergistic anticancer activity through the generation of reactive oxygen species (ROS), mitochondrial damage, and genotoxicity. Also, the NCs are more potent in inhibiting cancer cell growth than cisplatin, and their impact is unaltered in the presence or absence of TE pretreatment of A549 cells. The present study holds significant potential for various theranostic applications, which are highly desired for laparoscopic image-guided lung cancer therapy.

Bioinspired Photoresponsive Algosomes as a Nanocarrier for Combating Cancer and Bacterial Infections

The interplay between cancer and bacteria is a complex and evolving area of study, with certain bacteria implicated in both cancer onset and elicitation of chemoresistance. Combining nanoparticle-based photodynamic therapy (PDT) with antibiotics is a powerful strategy to overcome bacteria-induced chemoresistance in tumors. This synergistic approach enhances targeting and killing of both cancer cells and bacteria with a prompting immune response. In this study, we have synthesized uniform spherical shaped, self-assembled chlorophyll-rich, fluorescent, and photosensitive nanocarriers (named algosomes), derived from cyanobacterium, Spirulina platensis. These algosomes specifically target cancer cells due to an abundance of polyunsaturated fatty acids (PUFA) which are taken up more by the cancer cells to maintain cell membrane phospholipids to support their rapid growth and survival and exhibit tumor suppression via PDT due to integrated chlorophyll. Additionally, they serve as an excellent drug delivery system for antibiotics, enhancing the antibacterial activity against multidrug-resistant bacteria. Comprehensive biocompatibility evaluations on L929 cell lines, hemocompatibility tests, and in ovo biocompatibility studies in zebrafish embryos validated the safety of algosomes for potential clinical applications. In vitro findings demonstrated that algosomes in response to near-infrared (NIR) light kill cancer cells by inducing oxidative stress and mitochondrial damage, which leads to apoptosis in skin and colon cancer. Photoresponsive algosomes also impede cell migration following treatment, a crucial factor in preventing metastasis. Furthermore, the antibacterial and antihemolytic effect of antibiotic-loaded algosomes against multidrug resistant Escherichia coli and Staphylococcus aureus was quantified. This nanocarrier addresses the complex interplay of bacterial infections and cancer within a single nanoplatform, offering a promising solution to combat these dual challenges. Continued research and clinical development of this combination therapy hold promise for more effective and comprehensive cancer treatments.
