Corn Leaf Isoperoxidase Reaction to Mechanical Injury and Infection with Helminthosporium maydis

Abstract: Mechanical injury or infection with Helmintosporium maydis race T or O en proxidase acti in leaves of two corn ibreds which differ in their susceptibility to the fungal race T. Increases in act were found in the soluble fraction extracted from tissues with 20 mm phosphate buffer (pH 6), and in the ionicafUy bound fraction extracted fromn wail debris with 0.6 to 1 M NaCa; the covalently bound wai peroxidase fractin was unaffected. Mechanical injury and infection with either race enhanced the same distinctive… Show more

“…Repeated freezing and thawing, sonication, and maceration with glass beads was applied during extraction at 0 to 4°C. The enzymes were assayed as described previously (3,4) in the presence of 1 mM MgCl2 (14). The enzyme inhibitors DFMO (DL-a-difluoromethylornithine hydrochloride monohydrate, MDL 71782) and DFMA (DL-a-difluoromethylarginine hydrochloride monohydrate, MDL 71897) were synthesized at Merrell Dow Research Institute.…”
