Correlative biochemical and morphological studies of myelination in human ontogenesis

Abstract: Biochemical, light and electron microscopic observations in six human fetuses between the 16th and 34th weeks of gestation and five infants, 1 day to 3 years old, are presented. The results indicate that myelination of the human spinal cord started before the 16th week of gestation, as a considerable amount of myelin is isolated at this time biochemically, and occasionally axons with loose myelin coils are observed in the electron microscope. It is also stressed that morphological studies are insufficient to e… Show more

“…Double-label immunohistochemistry was initially performed on embryonic rat dorsal roots, since Schwann cells in these nerves are among the first to differentiate during embryogenesis (Niebroj-Dobosz et al, 1980). SCIP was first detected in the nuclei of LNGF-R 1 S-100 1 Schwann cells in the dorsal root proximal to the spinal cord on E17 (Figs.…”

“…However, myelination is not a postnatal process in every species. For example, in the chicken, MBP is first detected at 13-14 days of incubation (Hartman et al, 1979; Macklin and Weill, 1985); in human spinal cord, myelin can be isolated from fetuses at 16 weeks of gestation (Niebroj-Dobosz et al, 1980), and at that time, MBP has been observed by electroimmunoblotting (Roth et al, 1987).…”

“…Myelin abnormalities were found in 1074 the spinal cord of SOD1 transgenic rats progressing with the 1075 evolution of symptoms (Niebroj-Dobosz et al, 2007), and an 1076 extensive degeneration of gray matter oligodendrocytes in the 1077 spinal cord of mSOD1 G93A mice was reported prior to disease onset 1078 (Kang et al, 2013). Moreover, they showed that selective removal 1079 of mSOD1 from oligodendroglia substantially delayed disease 1080 onset and prolonged survival.…”
