Tooth decay or dental caries is an important oral health problem involving people of all age groups. The disease is the outcome of the demineralize process in which aciduric and acidogenic bacteria in a biofilm decompose tooth structure. Dental sealant, a resin material, which is applied on the occlusal pit and fissure surfaces of the teeth as a protective layer has been commonly used to prevent dental caries. However, the microbial effect on food residue is found to be a major cause of microleakage of sealant and secondary caries. Several types of antimicrobial agents were introduced to increase the caries preventive effect of dental sealants. Vanillin, the main component of flavoring agent vanilla, has been found to have antimicrobial property against Gram-positive and Gram-negative bacteria. The objective of this study was to investigate the antimicrobial effect of vanillin-incorporated dental sealant against biofilm formation of cariogenic bacteria. Dental sealant resin samples (Clinpro; 3M ESPE, USA) were prepared in 96-well plate in accordance with the amount of vanillin adding (0%, 0.5%, 1% and 5% vanillin). The cariogenic bacterial suspensions of Streptococcus mutans ATCC 25175 and Lactobacillus casei ATCC 334 were added to saliva-coated samples and incubated at 37°C in 5% CO2 atmosphere for 48 h to allow the biofilm formation. The quantity of vital biofilm was determined by WST Microbial Cell Counting Kit (Dojindo Molecular Technologies, USA) at 460 nm. One-way ANOVA and Tukey’s test were applied to the statistical analysis. A significant inhibitory effect against L. casei biofilm was observed in all vanillin incorporated samples (0.5%, 1% and 5% vanillin) compared with samples without vanillin. The percentage of biofilm reduction was 32-39%. For S. mutans, the suppressive effect was noticed only at >1% vanillin with 18-25% biofilm reduction. In conclusion, the incorporation of vanillin to dental sealants could decrease biofilm formation of cariogenic bacteria (S. mutans and L. casei). The use of dental sealants containing vanillin could be a promising measure to prevent dental caries due to their antibacterial biofilm formation property.

Berberine (BBR), a traditional Chinese phytomedicine extracted from various parts of Berberis plants, is an isoquinoline alkaloid used for centuries to treat diabetes, hypercholesterolemia, hypertension, and so forth. It has recently received immense attention worldwide to treat cancer due to its potent pro-apoptotic, antiproliferative, and anti-inflammatory properties. BBR efficiently induces tumor apoptosis, replicative quiescence and abrogates cell proliferation, epithelial-mesenchymal transition, tumor neovascularization, and metastasis by modulating diverse molecular and cell signaling pathways. Furthermore, BBR could also reverse drug resistance, make tumor cells sensitive to current cancer treatment and significantly minimize the harmful side effects of cytotoxic therapies. This review comprehensively analyzed the pharmacological effects of BBR against the development, growth, progression, metastasis, and therapy resistance in wide varieties of cancer. Also, it critically discusses the significant limitations behind the development of BBR into pharmaceuticals to treat cancer and the future research directions to overcome these limitations.

Background. Coronary heart disease (CHD) is the leading cause of death and disability worldwide. Accumulating evidence reveals that atherosclerosis (AS), characterized by systemic, chronic, and multifocal disease, and is the primary pathological basis of cardiovascular diseases, including CHD. However, the molecular underpinnings of CHD are still far from well understood. Our study attempted to identify aberrant plasma exosome-derived circRNAs and key exosomal circRNA biomarkers for CHD. Methods. The expression profiles of mRNAs, circRNAs, and lncRNAs in the blood exosomes of CHD patients and healthy controls were obtained from the exoRBase database. The corresponding miRNAs of the differentially expressed mRNAs, circRNAs, and lncRNAs were predicted via ENCORI and the miRcode database. LncRNAs/circRNAs and mRNAs with the cotargeted miRNAs were selected to construct an interaction network. Multiple machine learning algorithms have been used to explore potential biomarkers, followed by verification in patients with CHD using real-time quantitative reverse transcription-polymerase chain reaction (RT-qPCR). Results. Based on the cutoff criterion of P<0.05, we identified 85 differentially expressed circRNAs (4 upregulated and 81 downregulated), 43 differentially expressed lncRNAs (24 upregulated and 19 downregulated), and 312 differentially expressed mRNAs (55 upregulated and 257 downregulated). Functional enrichment analysis revealed that the differentially expressed mRNAs were involved mainly in neutrophil extracellular trap (NET) formation and the nucleotide-binding oligomerization domain- (NOD-) like receptor signaling pathway. Further analysis revealed that the DEGs in the circRNA/lncRNA-miRNA-mRNA interaction network were closely related to lipid and atherosclerotic signaling pathways. Hsa_circ_0001360 and hsa_circ_0000038 were identified as potential biomarkers for CHD based on three machine learning algorithms. The relative expression levels of hsa_circ_0001360 and hsa_circ_0000038 were significantly altered in plasma exosomes from patients with CHD. ROC curve analysis revealed that the areas under the curve (AUCs) were 0.860, 0.870, and 0.940 for hsa_circ_0001360, hsa_circ_0000038, and the two-gene combination, respectively. Conclusion. The circRNA/lncRNA-miRNA-mRNA interaction network might help to elucidate the pathogenesis of CHD. Hsa_circ_0001360 combined with hsa_circ_0000038 might be an important diagnostic biomarker.
